| Houttuyfonate,the decanoyl acetaldehyde,was a chemical component in the volatile oil of Herba Houttuynia(known as Zhe Ergen)which showed the inhibitit pathogenic microorganism.Sodium houttuyfonate(SH)was the affixture of sodium bisulfitewith Houttuyfonate.Wang D.(2002)reported that the level of IgG and IgM in the serum of the mice increased when the mice was injected with the bovin serum albumin(BSA)or red blood cell(RBC)mixed SH.Acording to the results,Wang D suggested that SH is an immunoadjuvant.SH has the effection of increasing the phagocytosis of macrophages and their respiratory burst and the concentration of free calcium in the cells and promoting T cells for excreting IL-2.Whether SH has or not enhance Avian immunity? No any reference was founded so far.It were reported that SH increase the antibody titre in the serum of chicken aginst the BSA antigen and protetion rate of duck immunized with inactivated bactrin.1 BSA antigen,mixed with SH(SH-C10)or mineral oil adjuvant,was injected to chick twice and the antibody titre was detected bu indirect ELISA.It was found that the antibody titers in the groups of BSA-C10and BSA-MO were significantly higher than that injected BSA only group(P<0.01).The antibody level aginst BSA between BSA-C10group and BSA-MO group was not significant(P>0.05).The results indicated that SH has the enhancement effect of antibody of was the same as mineral oil adjuvant after added into BSA immunity chick.In the second test,by immunizing ducks with inactivated Riemerella anatipestifer(RA)as antigen,we examine the adjuvant effectiveness of SH to bacteroidal particulate antigen by artificial infection.Lauroyl acetaldehyde sodium sulfite(SH-C12)as test character,AF serotype of RA as test strain to prepare inactivated Vaccine(iRA),the rate of SH-C12with iRA,the times of immunization and age of first immunization were filtered by orthogonal experiments.It showed that if the admixture of RA and SH-C12 was immunized on a time in ducks of 4 day-old,regardless 6 day-old,then after 11 to 15 days' immunizing,the rate of relative protection(RP)were below 40%.Reversely,RP of 5 groups of admixture of RA and SH-C12were greater than 60%when those immunized one time in 2 day-old,4 day-old,6 day-old,or 8 day-old,then secondary immunity after interval 5 to 7 days,RP of individual compounding group even reached 100%and both incidence rate and mortality rate were 0.The results indicated that: SH-C12added to iRA,the adjuvant effectiveness was fine after twice immunity.Finally,we selected relative more effectiven combination group of SH-C12and iRA to carry out ??experiment,compared with mineral oil(MO)adjuvant.4 day-old ducks were immunized for the first time and boostered when 11 days old,then challenged in the 16 days old.It found that the mortality rate of challenge control group was 53.3%(8/15),the incidence rate and mortality rate of SH-C12and MO groups both were 0 and their RP also were 100%(15/15).Compared with increasing weight on the seventh day after challenged,the average increasing weight of MO groups was lower than that of health control group,reversely,that of SH-C12group was higher than that of health control group.It showed that there were not disparity.In the second test,we found that the mortality rate of SH-C12control group(only injectedSH-C12 solution)was 81.8%(9/11),higher than the mortality rate(63.6%,7/11)of AF serotype RA challenged control group.So we examined accredited decanoyl acetaldehyde sodium sulfite(SH-C10)and Lauroyl acetaldehyde sodium sulfite(SH-C12)by qualitative and quantitive detection in accordance with Lauroyl acetaldehyde sodium sulfite sodium sutfite(SH-C12)standard substance on the basis of "the People's Republic of China pharmacopeia" 2000 edition second part.It showed that according to the items of FeCl3 response,I2 response,DNP-hydrazine response and ultraviolet spectrogram,SH-C10was consistent with the standardization of "pharmacopeia",and the content was 97.25%.FeCl3 response of SH-C12(inspect the hydroxyl)was negative,and others all accorded with "pharmacopeia",its content was 94.23%.To comprehened the toxicity of SH to ducks,intramuscular injection SH into 6 days old ducks lmg per one. The duck's growth and development were fine and were not find abnormal pathological change.It was reported that SH could cause haemolysis of human,but whether SH can cause haemolysis of ducks was not reported until now.So health duck blood was collected to determine haemolysis degree by spectrophotometric method in vitro.It indicated that 3 kinds of SH samples,including standard substance,its concentration ranging from 40μg/mL to 200μg/mL could make obvious haemolysis effect,and the rate of hemolysis was 96.0%.From RBC smear that added SH,we could see a lot of RBC only with nucl-eus but with no endochylema observed by Microscope.solution included Tween-80,when its concentration the same as SH,it could cause RBC of ducks to dissolve,but the highest rateof hemolysis was only 30.1%.It indicated that:RBC of ducks were seriously dissolved by SH,which maybe was the cause of increasing sensitivity of RA attacking ducks due to SH used alone.Above all,SH have the adjuvant effect that can be studied and empoldered.But utility dosage of SH and SH causing RBC of ducks to dissolve should be paid more attention and deep studied. |
