Xiangxi yellow cattle were analyzed by using of six microsatellite markers inthis study, the SAS software GLM process was used for analyzing relationshipbetween microsatellite markers, its alleles and body length, chest girth, etc. Withheterozygosity, polymorphism information content (PIC) and other parameters,genetic diversity of Xiangxi yellow cattle was analyzed. Furthermore, clusteringamong Xiangxi yellow cattle, Luxi cattle, Qinchuan cattle, Nanyang cattle, Jinnancattle and Yanbian cattle was proceed according to Nei's genetic distance. The resultsshow as fellows:1. The relationship between microsatellite IDVGA27 and chest girth, deepness ofchest was significant (P<0.05), and its allele 144bp had the most positive effects.There was a significant relationship between ETH10, BM2113 and deepness ofchest(P<0.05), and allele 248bp, 158bp had the most positive effects, respectively.The relationship between BM2113 and waist width was significant(P<0.05), and themost positive effective allele was 158bp. There were significant differences betweenBM2113, BM1824 and height at hip cross(P<0.05), and their allele 141bp and 169bphad positive effect.2. There were 65 alleles was detected at 6 loci. Average per loci was 9.3. Theallele number of ETH10 was 13 at best. The least was BM2113, and the number was 7.The average number of effective allele was 5.6583~11.1312, 7.8759. The number ofobserved allele was more than effective allele number. Distribution of allele wassymmetrical in the population of Xiangxi yellow cattle.3. Observed heterozygosity ranged from 0.8231 to 0.9102, and average was0.8669. Expected heterozygosity range from 0.8432~0.9175,and average was 0.8829.All microsatellite DNA were high polymorphism, and polymorphism informationcontent(PIC) ranged from 0.7998~0.9032, 0.8297 averagely. The results revealedthat six loci were adapted to analyze genetic diversity of Xiangxi yellow cattle, andgenetic diversity of Xiangxi yellow cattle was abundant, and genetic potential washigh.4. Genetic differentiation within three sub-population of Xiangxi yellow cattle(estimated Fst) was 0.3670, and individual transferring was at large (Nm=0.4312). x~2fittness test showed that Xiangxi yellow cattle population was not in a state of Hardy-Weinberg equilibrium at six microsatellites.5. The 6 population were divided into 3 clusters based on Nei's genetic distance.The first cluster was Luxi cattle, Nanyang cattle, Qinchuan cattle and Jinnan cattle.Yanbian cattle and Xiangxi yellow cattle was grouped solely, respectively.
