It was named by the small molecular weight and high transport ratio in Polyacrylamide gel electrophoresis. The certain studies manifested that HMG mainly bind in the field of nucleosome in eukaryotic activity open gene. HMG builded up the important function in the course of maintainning the activity structure of chromatin, gene transcription and replication. HMG have three families named HMGA, HMGB and HMGN. High Mobility Group Chromosal protein N2 is one menber of HMGN.Recent studies showed that people's HMGN2 had Antimicrobial activity in addition to the gene transcription regulation. Although HMGN2 have higher conservatism among species relatively, there are no relevant reports about Antimicrobial activity of HMGN2 from animals.The assay had HMGN2 gene from cattle rebuilt prokaryotic express plasmid pET-32a(+)-HMGN2 induced by IPTG, the refused protein with the Histidine-tagged was purified by His-Trap Kit and identified by SDS-PAGE. The antimicrobial activity was detected by agar diffusion test. The result is that the recombinant HMGN2 protein was against the pathogenic strains E.coli ML-35P and Staphylococcus aureus ATCC25923. The Antimicrobial activity is more obvious when the concentration is increasing. |
