| In this paper the partial sequences of ACCa gene from liver tissues of Landes goose, Tianfu meat goose and super meat duck were amplicated by RT-PCR, The ACCa gene was analyzed for Sequence variance and structure characteristic, protein speciality, the phylogenetic realitionships in different species of ACCa gene was analyzed, and the molecular evolution model of ACC, PCC, PYC and MCC genes which were determined from Biotin Carboxylase gene family by bioinformatics methods. These exprimental results showed:The ACCa gene sequences(669bp) of Landes goose, Tianfu meat goose and super meat duck were received, which coded 188 amino acid. The similarity analogical percentage of nucleotide sequences during between Tian fu meat goose, and Landes goose, duck, chicken, human and house mouse were 100%, 99.1%, 92.0%, 82.9% and 82.7% respectively, while amino acid similarity were more than 92%.At the same time, the received ACCa gene sequences were compared with chicken sequence from GenBank(GenBank sequcece number: NM205505), 38 variable sites were found, including 34 Singleton variable sites, 4 Parsimony informative sites, but no three variants and four variants were found, and nucleotide diversity was 0.02980.As far as average content Base composition of partial sequences(669bp) of ACCa gene are concerned, the differences from three birds in the same base are Small, the content of codons first, second, third during different bases are in different degree, the variances scope from the third location were much higher than the first and second codon location.All the probability of transition (si) was higher than that of tranversion (sv)(si/sv =2.0~3.5) between the goose and duck, duck and chicken, duck and goose, and the transition were 28, 26, 4 respectively, while the tranversion were 8, 8, 2 respectively. For the codon location, besides there was one transition in the first and second codon location between duck and chicken, goose and chicken respectively, the other transitions present to the third codon location. No tranversion was found between the first and second coden location. The total number of synonymous sites (SynPos) between duck and chicken, goose and chicken (155.00—157.83) were far lower than the total number of nonsynonymous sites(NsynPos)(504.83—511.17), differences was small during different species; The number of nonsynonymous differences (Ka) between duck and chicken, goose and chicken were both 0.0020, but no NsynPos was found between duck and goose. The number of synonymous differences(Ks) was 125.05—134.35 times to the number of nonsynonymous differences(Ka).From the Neighbour-Joining tree of ACCa gene, the Landes goose and Tianfu meat goose were first clustered in one branch, and then clustered with duck, then clustered with chicken. The two mammal human and house mouse clustered in one branch alone. Thus, two branches, Aves and mammal were formed.Analysis of ATP grasp protein Domain of the ACC1, ACC2, PYC, MCC1 gene(sub-gene) which were determined from members of Biotin Carboxylase gene family indicated that, conservative ATP binding site, Carbamoyl-phosphate synthase subdomain signature 1(CPSASE1) and specific structure of Biotin Carboxylase gene family GGGGKG (GGGGRG) segment were found in this domain.Furthermore, the phylogenetic analysis indicated that three branches were formed in the Biotin Carboxylase family gene including ACC, PCC2, PYC, MCClgene, which were ACC branch, PYC and MCC branch, PCC2 and MCC2 branch. The duck, goose ACC1 and chicken ACClwere clustered in one branch, formed ACC1 branch of Ayes(Bootstrap =99%), then chustered with ACC1 branch of mammal(Bootstrap=99%), then chustered with ACC2, ACC branch of BC family were formed; the PCC2 sub-gene of different species chustered with PCC2 branch, then clustered with MCC2 (BP=100%), PYC clustered with MCC1 (BP=77%), those indicated that there were later, closer common ancestor between PCC2 and MCC2, PYC and MCC1 respectively. |
