Study On Development Of Grain And Differences Expression Of MRNA In Triticum Aestivum Under Different Concentration Of Zinc In Vitro Cultivatio | | Posted on:2008-01-27 | Degree:Master | Type:Thesis | | Country:China | Candidate:X J Dong | Full Text:PDF | | GTID:2143360215995514 | Subject:Crop Genetics and Breeding | | Abstract/Summary: | PDF Full Text Request | | Zinc is the necessary nutrition factor for plants, human being and animals.So it goes by the name of anima element.It bears many physiology fuoction.It isn′t only the chemical composition of protein essential elements, but also is the composition of chlorophyll, enzymes and ribose.In life-sustaining activities and increase crop yields, improve quality of products, it takes a crucial role.The formation of wheat grain yield and quality is closely related to zinc nutrition.In order to definitude the accumulation of zinc in different organ of wheat,and discuss the regulate effect of zinc on agriculture character, grain protein and different expression of mRNA in wheat after anthesis, we studied the law of absorption and utilization of zinc ,the development laws and accumulation dynamics of high molecular weight-glutin subunit(HMW-GS), different expression of hordein and gene in Xinchun8 and Dulun under different zinc concentration by SDS-PAGE ,A-PAGE electrophoresis and DDRT-PCR techniques in vitro cultivation. The results showed that:(1) Transformation trend of zinc content was basically consistent in different organ except under high concentration of zinc.The relation of content of zinc among different organ was ear>the first joint under spike>the second joint under spike.(2) The growth of wheat was restrained for lack of zinc,especially the elongate of joints was greatly suffocated.The joint length of Dulun was shorter than that of Xinchun8.The effect of zinc on length of joints in Dulun is more evident than that in Xinchun8.The first joint under spike reacted to zinc more sensitively than the second joint.Zinc slightly affected on chlorophyll content in flag leaf at anthesis,but greatly affected on it two weeks after anthesis.(3) Zinc evidently increased the grain number and percentage of wheat.Tiny zinc consumingly restrained the floweret polarization and reduced grain number of Dulun.Superfluous zinc accelerated the floweret polarization but reduced grain percentage.Grouting of grain was counterworked and 1000-grain weight was reduced greatly for lack of zinc.The 1000-grain weight of wheat also reduced slightly under superfluous zinc condition.(4) Protein content of grain was not prominently affected by zinc.The protein component in wheat grain was prominently affected by zinc.The relationship of different protein component to zinc was discriminating with each other.(5) Expression period of subunits were greatly affected by concentration of zinc. With the accretion of zinc concentration, expression period of subunits brought forward. There was obvious difference in subunits compositions and expression period of subunits under the same concentration of zinc between the tow varieties.(6) There was some difference in gliadin protein compositions between Xinchun8 and Dulun. The difference was mainly inωzone,γzone andβzone, Difference inαzone was not apparent. The influence by the concentration of zinc ion on the two varieties was basicly consistent with each other, that is to say: gliadin protein was a little expressed in CK and low concentration of zinc ion while its expression was enhanced under other concentrations and expression quantification was almost the same.(7) Gene expression was affected by concentration of zinc in three ways: especial expression, enhancement expression and attenuation expression. 383 difference segments were identified in the two varieties during the experiment, there were 212 difference segments in Xinchun8 and 171 ones in Dulun. | | Keywords/Search Tags: | wheat, in vitro culture, zinc, agriculture character, protein, different expression of mRNA | PDF Full Text Request | Related items |
