1 The avUCP mRNA expression, the energy and fat metabolity of avian UCP transgenic chickensThe experimental chickens were chosen from one hundred and sixty avian UCP transgenic chickens and non-avian UCP transgenic chickens of 28 days after DNA and RNA detecting, and divided into two groups after weighting, avian UCP transgenic chickens were for experiment, non-avian UCP transgenic chickens were for control, beginning with growing perfonnance and metabolism experiment for 21 days. The experimental chickens were weighted at the age of 49 and 70 day, ten chickens of every group were killed for detecting partial carcass parameters.The result indicated that the avian UCP mRNA content of transgenic chickens was fifteen times as great as that of non-transgenic chickens, the interference of avian UCP gene restrained the expression of avUCP mRNA.The feed conversion, average dayly gain , food intake ,energy appearance metabolity ,crude fat appearance metabolity, energy true metabolity and crude fat true metabolity were different significantly between the transgenic and non-transgenic chickens,the ages and genders affected the growing perfonnance ,energy metabolity, fat metabolity and partial carcass parameters of avUCP transgenic chickens, with apparent amelioration.2 The effect of dietary nutrition structure on energy and fat metabolity of avUCP transgenic chickens2×4 two -factor variance analysis experiment was designed ,including avian UCP transgenic chickens and avian UCP non- transgenic chickens ,three diets (high energy diet, high fat diet, standard diet)and fasting group,consisting of 8 treatments. The experiment last for 28 days except the fasting group for 6 days, when the experiment ended, all chickens were killed to separate skeletal muscle for assaying ATP content by Bioluminescence and avian UCP mRNA expression level by real time PCR,detecting blood glucose,free fat acid,glyceride,cholesterol and insulin.The result were shown, the dietary nutrition structure and avian UCP gene affected the blood glucose, free fat acid, glyceride, cholesterol and insulin significantly (P<0.05) .The dietary nutrition structure affected avian UCP gene expression distinctly (P<0.01) ,high energy feed and high fat feed could fortify the avian UCP gene expression. The dietary nutrition structure and avian UCP gene affected the abdomen fat significantly. Furthennore, dietary nutrition structure affected the subcutaneous fat thickness distinctly.Avian UCP gene , the dietary nutrition structure and feeding period,all could significantly affect the skeletal ATP content, the skeletal muscle ATP contents of avian UCP transgenic chickens were higher than those of non-transgenic chickens among all dietary groups, furthennore, avian UCP gene and the dietary nutrition structure, the dietary nutrition structure and feeding period ,both had distinct interaction.
