There were 192 NewYangzhou Chicken and 120 Wenchang Chicken used in this experiment. In this study, the egg quality of New Yangzhou Chicken at the age of 40-weeks and the age of 60–week, the egg quality of Wenchang Chicken at the age of 60-weeks were estimated, respectively.The polymorphisms of 20 microsatellite loci on 2,4 and Z chromosomes from the two chicken populations were analyzed. The associations among three indexes of egg quality (HU,EW,EST) and 20 microsatellite marks were also analyzed by SPSS procedure .1 EW, ESW, EST, ES, HU, ESI, YW, YC of the egg of New Yangzhou Chicken at 40wk were 47.56g,6.24g,321.92μm,4.71kg/cm2,75.42,1.37,15.34,10.36, respectively. EW, ESW, EST, ES, HU, ESI, YW, YC of the egg of New Yangzhou Chicken at 60wk were 47.60g,5.76g,289.00um,3.34 kg/cm2,72.39,1.38,16.14g,9.95, resepectively. EW, ESW, EST, ES, HU, ESI, YW, YC of the egg of New Yangzhou Chicken at 60wk were 49.00g,6.34g,320.69μm,4.01 kg/cm2,70.02,1.31,16.45g,6.13, respectively.2 The correlations between each index of egg quality of Wenchang Chicken at 60 wk, New Yangzhou Chicken at 40 wk and 60 wk were analyzed by SPSS procedure. The significant correlations were for between EST and ES, between ESW and EST, between EW and ESW.3 Number of alleles of each Loci. in New Yangzhou Chicken and Wenchang Chicken were calculated ,and frenquecies of alleles and genotypes were accounted according to gene frequencies of 20 microsatellites. The average values of alleles number ,genotype, average PIC ,and heterozygosity of 20 microsatellite loci on 2,4 and Z chromosomes were 4.62(2-9), 0.599 and 0.521 in New Yangzhou Chicken. The average values of alleles number ,genotype, average PIC ,and heterozygosity of 20 microsatellite loci were 4.2(3-7), 0.601 and 0.622 in Wenchang Chicken.4 20 Microsatellite DNA primers were used to analyze whether there were some quantitative trait loci or main gene affecting partial egg quality traits in New Yangzhou Chicken and Wenchang Chicken populations. The results indicated that in New Yangzhou Chicken, haugh units at 40 wk for least squares means in ADL0217 and ADL0260,egg weight at 40 wk for least squares means in ADL0176, haugh units at 60 wk for least squares means in ADL0260 and MCW0239, egg weight at 60 wk for least squares means in ADL0176 and MCW0239, egg strength at 60 wk for least squares means in LEI0229 had significant difference; In Wenchang Chicken at 60 wk, haugh units for least squares means in ADL0176 and MCW0240, egg weight for least squares means in LEI0119, egg strength for least squares means in MCW0246 and LEI0229 had significant difference.
