The explosion of world population spurs a need for more food supply. Thepopularization of hybrid varieties plays a major role in the increase of crops productions in the past few decades. Although utilization of heterosis has achieved great success, the mechanism underlying this phenomenon remains poorly understand. In turn, it limits the full utilization of heterosis. This dilemma plus the environmental stresses and disasters urgently call for further understanding of heterosis. It is important to isolates the genes differentlyexpressed in hybrid and parental lines, because it is necessary for understanding.In our experiment,We investigated the expression patterns of 10-42,10-44,13-42,13-44 to analyse the different expression in addition and loss alien chromosomes of wheat-wheatgrass by cDNA-AFLP, isolated a large number of differentially expressed genes and confirmated some of them.The results are listed below:1. The wheat leaves were taken as the material, used the cDNA-AFLP method to analyze the gene expression characteristics between two kinds of samples, and separated the different expression genes between them.256 combinations of AFLP primers produced about 600 differently expressed fragments, The different expression patterns were present/absent and quantity , of which 269 specific fragments were cloned and sequenced and their function sought in the genbank database.Blast (EST) comparision indicated the result that majority of them come from Triticum cDNA library, because the research to the wheat gene function is not enough, the accurate function of most genes had not been found.2. The sequences were analysed , 79 differences fragments were chosed to design specific primers to perform the RT-PCR and Real-Time PCR examination.The results indicated 29 genes were differently expressed, 11 of them were present/absent , 18 of them were quantity difference .One of the present/absent different expression genes was caused by the different wheat genome.3.Besides the genome difference, methylation may also had affection on differenr gene expression, 9 of the present/absent genes were choosen to design methylation specific primers. compared The sequences indicated that the methylation had the certain relationships to gene expression, but if it was the direct cause of the different expression is not clear. There may also be many other regulative mechanism, gene expression was regulated by many kinds of mechanism.
