A study is about the different breeding methods in Oriental hybrids (Sorbonne),(Acapulca); Lilium longiflorum (Sayaka) and Asiatic hybrids (High class),(Golden horn). Making researches on overcoming the incompatibility of Lilium. The purpose is to observe the growth process of pollen tube in the different lilium variety and investigate the efficiency of overcoming the incompatibility by the different breeding methods in lilium breeding, at the same time research on the growth process of Lilium microspore. The result of the cut-style pollination experiments shows that the stigma has no any restrain effect on the pollen bourgeon in Lilium. The preceding fertilization barrier substances of"Sorbonne"×"Sayaka"disperse in the style equally, and the substances of"Sorbonne"×"High class"mainly exist in the combining part of stigma and style. If cutting the upper middle part to style and pollinating, all the pollen tube can grow rapidly. Cutting style of different length and then comparing the methods, you will find that style which is remained 0.5cm is the most useful for overcoming the preceding fertilization barrier. The pollen tube extends along the style. The amount of pollen tube decreases gradually according to stigma, style, ovary. The amount of pollen tube in the stigma> the amount of pollen tube in the style> the amount of pollen tube finally in the ovary. Two months later, the result of the embryo rescue method,"Sorbonne"×"Acapulca"and"Sorbonne"×"Sayaka"all can get seeds, but cut-style pollination methods can't get more seeds as normal stigma pollination method in"Sorbonne"×"Acapulca"; on the contrary, normal pollination on the stigma can get more seeds than cut-style pollination methods in"Sorbonne"×"Sayaka"."Sorbonne"×"High class"can't get seeds in the end. It is because the types of genes of parents'lines are really different, sperm cells and ovum cells can't conjoin and form hybrids. The pollination method in vitro can overcome the incompatibility and get the hybrids. The cut-style reated with pollen culture solution can accelerate the growth of pollen tube, but can not overcome the incompatibility of Lilium.The results of experiment on hormone manage show that hormone can accelerate the growth of the pollen tube, no matter it is self-pollination or cross–pollination. 5 hormones stimulate action ranking is BA>GA>NAA>IAA>IBA; the action time for 5 hormones stimulate to the pollen tube growth of self-pollination and cross-fertilize is different. For self-pollination, 5 hormones stimulate action is at the prophase of the growth of pollen tube, however, for cross-fertilize, 5 hormones stimulate action is at the anaphase of the growth of pollen tube. GA can overcome the incompatibility in self-pollination"Sorbonne", and gain one embryo seed; others can not gain embryo seed. The result of bud and delay pollination experiment shows that bourgeon quantity and growth speed of pollen in different mature degree of stigma and style is different. When pistil becomes more mature, the pollen bourgeon can get more quantity and the growth of pollen tube is faster. If bud pollination can't overcome the incompatibility in self-pollination and cross-fertilize"Sorbonne", and it can not gain the seed; delaying 5d pollination can overcome the incompatibility in self-pollination"Sorbonne", and gain two embryo seeds. In delay pollination experiment,there is little difference between the growth of estate of the pollen tube in self-pollination, but in cross-fertilize, the growth of estate of the pollen tube will rise firstly and then fall。The result of Lilium microspore growth process experiment shows: the sequence of"Golden horn"microspore growth process is:-16d bud (16d before blossom), pollen mother cell phase; -15d~-13d bud, dyad phase; -13d~-11d bud, tetrad phase; -10d~-9d bud, 1-cell pollen(can cultivate microspore); -9d~-6d bud, 2-cell pollen. In -5d bud, pollen has already grown into mature pollen, it can pollinate , along with bud growth , bourgeon rate will be increased according to pollination, the best of all is from -1d and 0d (intraday blossom).
