| Bulblets of Oriental lily'Siberia'and'Sorbonne'were selected for in vitro culture. Three aspects of primary research were processed, that was, the formation and enlargement of in vitro bulblet; the characateristic of growth and dormancy and transplantation of bulblet in vitro. The results are as follows:1. The formation and enlargement of bulblet in vitroThe effects of the concentration of sugar, NAA, macroelement, light and comprehensive factors on bulbet's in vitro formation and enlargement were researched. 90 g/L sucrose is favorable for formation and enlargement of bulblet. Lower concentration of NAA can add the rate of bulblet's formation. It was favorable for bulblet's formation and enlargement as increase the concentration of macroelement. 2 MS was proved the best concentration. In dark the number and volume of in vitro bulblet were increased, but the number and size of leaf were inhibited. At last we found the MS medium with double macroelement, 0.2mg/L NAA, 90 g/L sucrose and dark was best for bulblet.2. The growth and dormancy of in vitro bulbletThe growth of bulblet in vitro is not same, and the growth of every part is not same too. The enlargement of bulblet is not unlimited. The soluble sugar content is high and starch content is low in plantlet before inducing bulblet, with the growth of the bulblet, the content of soluble sugar decresed, but the starch content changed contrarily. The bulblet in vitro has the characteristic of dormancy. Breaking dormancy need a certain low temperature, 4℃is better than 10℃, the bulblet of'Siberia'need 10 weeks 4℃low temperature for breaking dormancy, but'Sorbonne'need 8 weeks 4℃low temperature. Cold treatment with the whole plantlet is better than cold treatment without leaves.3. The transplantation of bulblet in vitroWhen the diameter of bulblet in vitro is bigger than 1.5cm, it is easy to survive if manage rightly after transplanting, generally the survival rate is 90%. Prelite / peat (2 /1) is the best medium to transplant. Fertilizing 1/2MS nutrition is favorable for the growth of plantlet. |
