Avian reovirus(ARV) infection exsit in the poultry industry around the world. ARVs can cause viral arthritis, tenosynovitis, malabsorption and chronic respiratory diseases, and it is also one of the immunosuppressive diseases and subsequently results in more susceptive to the infection of other pathogens, and cause serious economy loss for poultry industry. A lot of researchers are exploring the different diagnostic method and different vaccine of ARV in recent years.The construction of recombinant plasmid (pcDNA- σ3)with σ3 gene against Avian reovirus; the immunological efficacy of the recombinant plasmid (pcDNA- σ3); the cloning, expression and sequence of nonstructural protein gene σ NS of ARV are studies in the thesis.The σ 3 gene of Avian reovirus was amplified with the specific primers by RT-PCR. The purified RT-PCR product was inserted into pcDNA3.1(+) vector. The recombinant plasmid was identified by restriction endonuclease analysis and PCR. It was proved by DNA sequencing that the acquired recombinant plasmid contains complete σ 3 gene and the recombinant DNA vaccine plasmid of pcDNA- σ 3 was constructed .Three kinds of ARV vaccines pcDNA- σ 3 DNA vaccine , inactivated...
