The experiment succeed in detecting specific antibody and general antibody in serum, tear, tracheal and Duodenal irrigation of chickens IBV-infected from 1st day to 28th day and the control group by indirect ELISA which is fast, sensitive, better specificity and repeatability. Meanwhile, on the basis of strong adhesion capability and quick adherence, the experiment succeed in purifying macrophages by different-speed adhesion, and in obtaining splenic macrophage and medullary macrophage of chickens by regulating adhesion cultural time and temperature, then in detecting the activity of macrophages by assaying phagocytic capacity to neutral red, MTT colorimetric assay, assaying quantification of AcP, NO and IL-1. The results indicate that:1. IBV specific antibody could be detected on the 3rd day in SPF chikens infected IBV,and topped on the 14th ,and the level of anti-IBV antibody kept several weeks; the local immunity emerged eariler than that of specific immune response, the level of non-specific antibody was lower than that in control group. It shows that the total humoral immunity degraded , but the specific humoral immunity enhanced, while the region humoral immunity emerged faster but transent, the humoral immunity of the whole body emerged slowly but persistence time longer.2. The activity of macrophage in SPF chickens infected IBV was clearly higher than that of the control group. In splenic macrophage, this situation happened between the 7th day and the 10th day; while in medullary macrophage,this situation happened between the 3rd day and the 10th day, the 7th day and the 21st day. All that showed the infection of IBV would further the activation of macrophage, enhanced the capability of phagocytosis, the quanity of ACP in cells increased, the activity of secretion and the capability of the atigen presentation were both enhanced, and took part in the main progress of infection. That was basically same to the varitation of the humoral immunity, but it can last longer than the specifical IgG.3. During the infection of IBV, the quanity of IL-1 increased in macrophage which showed immune function of macrophage was improved. After the infection of IBV, the effect of macrophage was significant. Through cytokine network and the effect of antigen presentation, macrophage associated with other immune cells and improved speccific immune response of SPF chickens.That hints thaoral immunity of chicken decreases after IBV infection, while macrophages energetically participate in immune response of IBV and play an important part in resisting IBV...
