Based on the enhanced linkage map of the sheep genome in chromosome 3, we selected out 14 microsatellites RM150, OarCP43,OarCP0034, ILSTS22, ILSTS0049, BMS1953 BMS1350, BMS1248, BMS1048, BMS0640, BMC1009, BM3501, BM2830, BL0004 of sheep for studying in Liang Shan semi-woolsheep. And we experimented with fluorescence primers, multiple polymerise chain reaction and ABI310 capillary electrophoresis for paternity testing in the group of 360 lambs borning from 2000 to 2003. The allele frequency distributions of the 14 STR loci were studied. The heterozygosity (H), discrimination power (DP), probability of exclusion (PE), the accumulative probability of exclusion and polymorphic information content (PIC) were calculated. Meanwhile, we corrected the errors of pedigree recording.Finally, we found the biological parents of 11 lambs without pedigree recording, and modify the false recording of 5 individuals. We found the significant identity between the analysis result and pedigree recording in the group born in 2003. The accumulative probability of exclusion is 0.999981 when parents are both unknown. If a parent is known, the accumulative probability of exclusion is 0.999999.All of the 14 STR loci have higher polymorphism and had high heterozygosities and PIC. The data on the allele frequencies of these 14 STR loci can be used in construction of molecular genetic map, Identification of sheep economic trait QTLs. Meanwhile, it is in favor of researching the group structure of sheeps, the diversity of heredity, individual identification, paternity testing and other population genetic researches.
