Six species of animal (chicken, goose, dog, rabbit, pig and goat) were used to study the sensitivity to soybean agglutinin(SBA). Two methods, cyto-agglutination and sorbent assay, were employed in the experiments to identify the influence of purified SBA on the animals. The agglutinating activity of SBA with normal red blood cell, trypsinized red blood cell and spleen lymphocyte were studied in the experiment of cyto-agglutination by means of obsorbent density(OD). The binding capacity of SBA to small intestine mucous membrane (SIMM) and whole tissue of small intestine (WTSI) were studied in the experiment of sorbent assay with the method of ELLSAPurified SBA was isolated through the linked guar-amylose.In the experiment of cyto-agglutination, concentration of SBA was planed with double-diluted method, and the OD of single-cell suspension was measured at 620nm after an effective period of agglutination. In the experiment of sorbent assay, SBA was subject to nine successive dilution grades with the same buffer and used as an antibody marked by horseradish-peroxidase(SBA-PO). Binding capacity of SBA to SIMM and SIWT was measured through the activity of peroxidase at OD 495nm.The results of cyto-agglutination of SBA with normal red blood cell showed that rabbit was the most sensitive to SBA; pig took the second place; goat was sensitive only in certain concentrations of SBA;chicken, goose and dog were none sensitive to SBA. As for rabbit, the OD was almost zero among the concentrations of SBA diluted by two to sixty-four times, where a strong agglutination existed. While there was no agglomeration when SBA was diluted by 1024 times, at 1.50×10-3mg/ml, which might be the critical concentration. There was strong agglomeration before 16 times dilution and no agglomeration at 128 times, 1.25×10-2mg/ml in pig. Goat was special, there was only weak agglomeration in the midst concentration of 2.52×10-20.201 mg/ml.The results of agglutination of SBA with trypsinized red blood cells showed that the sensitivity to SBA enhanced in some animals after trypsinization. Rabbit expressed four times of sensitivity at 4096 times dilution and a might be critical concentration of 0.39×10-3mg/rnl of SBA. The respective results in pig were six or eight times sensitivity, 1024 times dilution and 1.5×10-2mg/ml critical concentration. While there was faintish agglutination at two or four times dilution of SBA in chicken even though none before trypainization, the reason might be exposing of binding site to SBA then.
