| Tung-seed meal extracted by soaking, containing 36.29%-45% protein, and is a good source of plant protein feed. But the toxin in tung-seed meal, which is uncertain as yet, holdbacks the application of rung meal on animal production. The study was based on the previous researches to explore the toxin and active substances in tung-seed meal, isolate and purify the active elements of the meal. A crystalline purified was identified as lignan by UV, 1H-NMR, 13C-NMR, DETP for the first time. Furthermore, the physiological activities of extracts were studied.Tung seed meal was processed as follows: after crashing, the granules of the meal went through 40-hole sieve, then extracted by 95% ethanol with the ratio of meal: solvent = 10:1 at the extraction temperature of 80℃ for 2 h. The crude extract was enriched by the NKA macroporous resin, and the 30%-ethanol elution was purified with polyamide chromatographic column. Then HPTLC and HPLC were used to qualitatively analyze the fractions from Polyamide chromatographic column. A white needle-shaped crystalline was discovered. The colorless needle-shaped crystalline was characterized as (±)3,3'-bisdemethylpmoresinol by UV, 1H-NMR, 13C-NMR and DEPT spectrum.MTT colorimetric method was used to investigate the cytotoxical effects of water extract and ethanol extracts from Tung-seed meal on chick embryo fibroblast (CEF). To determine the antivirus activity of extracts on new castle disease virus (NDV) in vivo and in vitro, Tung-seed meal extracts were treated with NDV for 2 h at 37℃, then inoculated in 10-day chick embryo to measure EID50. After that, Tung-seed meal extracts treated with NDV were inoculated in 10-day chick embryo and CEF. The results suggested that the extracts with 70% ethanol elution via NKA resin had the most cytotoxicity, while water extract was the least cytotoxical. But the water extract was most effective in anti-NDV (0.125mg/ml) , then the anti-NDV capacity of the extracts with 30% ethanol elution via NKA resin (0.2mg/ml), water extract (0.5 mg/ml, 0.25 mg/ml), absolute alcohol extract (0.2mg/ml) decreased consequently. |
