The pig epidemic disease was a problem bothering the pig industry for a long time. The piggery in a poor environment was incentives for many pig diseases. It could reduce the occurrence of pig diseases and improve the ability of pig production by providing a comfortable, clean living environmental site. The microbial-fermentation bed was a kind of newly arisen pig-raising mode, which provided the pig-breeding industry with non-odor and zero-pollution. The biggest advantage in the pig-raising mode was to create a natural defense against the pig disease to improve the animals' immune level and reduces the incidence of mortality greatly.In this study, the diversity of microbial community structure and dynamics of pathogenic bacteria in the microbial-fermentated stroma cushion of piggery were investigated using phospholipids fatty acids (PLFAs). The results showed as follow:(1) As revealed in the results, the fermentated stroma cushion contained numorous microbials among which bacteria dominated far over fungi and actinomycetes.(2)The number of bacteria in the cussion increased at the beginning and then decreased with a mean of 1×10~8 cfu/g during the whole process. However, the numbers of fungi and actinomycetes were between 1×10~3 and 1×10~4 cfu/g. The bacteria isolated from the stroma cushion belonged to 4 kingdom, 22 genus.(3) Some amount of Escherichia coli and Salmonella enterica distributed in the stroma cushion, but for a period of time, the number of bacteria would continue to decline and remained at a very low level. It indicated that the stroma cushion could suppress the pig diseases.(4) Brevibacillus brevis which were used as a pilot bacterium could be planted in the fermentation bed and maintained as predominant bacterium which was tested to have antagonistic action on E. coli and S. enterica, which was evidenced by the suppression circle experiment that the brevibacillus bacterium had the obvious inhibitory action to E. coli and S. enterica. It meant that B. brevis played an important role in suppressing the pig diseases.(5) The PLFA biomarkers were introduced into analyzing the diversity of microbial community in the stroma cushion of pigsty. The result showed that 41 biomarkers were detected, which increased a rich microbial in the stroma cushion. Based the indexes of Shanon and pielou being analyzed, it was found that the value of Shanon and Pielou were not obviously changed among different using methods, which showed the microbiol community structures of stroma cushion were relatively stable.
