Tenebrio molitor, in the face of low temperatures, woμLd inhence the expression of antifreeze protein, however, there are many members in the antifreeze protein gene family, the length and structure of the members is different. It's not clear, which antifreeze protein gene expression higher in the adaptation process.In this article, we selected three representative genes whose conserved sequence is different in length from Tenebrio molitor antifreeze protein gene family in NCBI. They are AF160497, AF160496 and AF160494. We determined relative gene expression levels using realtime fluorescence quantitative PCR. Handling as follows, Tenebrio molitor were accli-mated at 4°C, 4 days, 8 days, 16 days and 24 days. We checked the expression of three genes. And we also carried out the Tenebrio mortality statistics of -11°C at 4°C cold acclimation 4 days, 8 days, 16 days and 24 days. In addition, we also carried out expression of antifreeze proteins in eukaryotic cells Sf-9.The resμLts showed that at 4°C with the Tenebrio treatment time, -11°C for treatment, the mortality rate found in Tenebrio shown a downward trend, 4 days, 8 days, 16 days had significantly reduced, and to the 24 days and 16 days, the difference was not significant.The gene of Af160497 acclimation at 4°C for 4 days, 8 days, 16 days and control, sampling of genes RTPCR. Expression of gene Af160497 time to deal with the 4-day expression of a large number of sampling points up to 8 days and the expression of less than 4 days, after 16 days to process after the gene expression continue to increase, the overall upward trend.The gene of AF160496 acclimation at 4°C for 4 days, 8 days, 16 days and control, sampling of the genes of the RTPCR. AF160496 gene expression levels in 4 days of samp-ling points have been greatly improved, but as time goes on, we in 8 days and 16 days of sampling reality, it is the expression remained low.The gene of AF160494 acclimation at 4°C for 0 days, 4 days, 8 days, 16 days, samp-ling the genes RTPCR. AF160494 gene expression in the 4-day sampling point is also been greatly improved, and the sampling points in the eighth day also showed lower gene expres-sion.By comparison of the three genes, the overall expression of genes AF160494 higher than the other two in a dominant position, level of AF160496, AF160497 is very, signify-cantly lower than the level of gene expression of AF160494. In this study, we examined the effects of the mortality of cold exposure on low temperature and gene expression of different types of antifreeze protein members in the family. The resμLts showed that cold acclimation can significantly reduce the mortality of the larva and improve expression of antifreeze protein gene. And, the longest antifreeze protein is most apparent to low-temperature-related morta-lity. It means that the function of different antifreeze protein family members has a clear division, and it provides a new understanding of the use of antifreeze proteins and antifreeze protein activity.
