| Virus infection can cause severe damage to the production in silkworm industry. Bombyx mori densonucleosis virus (BmDNV) is one of the principle pathogens causing flacherie disease in the silkworm. BmDNV is species-specific and can infect silkworm larval midgut columnar epithelial cells which then causing the death of silkworm. In our country, most of the Bombyx strains are highly susceptible, while other strains exhibit complete resistance to BmDNV infection. The two-hybrid system is a powerfull tool to detect and identify proteins that interact with a protein of interest. To gain a better understanding of the mechanisms underlying resistance to BmDNV-Z in some Bombyx strains, interaction between BmDNV-Z and silkworm larval midgut cells were screened in this study using yeast two-hybrid system.By the mate of yeast Y187 strain that have been transformed with recombinant prey vectors of seven ORFs of BmDNV-Z with silkworm larval midgut cDNA library, we identified twelve host midgut proteins that interact with BmDNV-Z including Bombyx mori 35kDa protease,hypothetical protein,Bombyx mori ribosomal protein S5,Bombyx mori transgelin,Bombyx mori serine protease precursor. |
