| Peanut is an important oil crop and cash crop.The infection of penut by aflatoxin of aspergillus flavus causes great influence on the production, processing,and trading of China. The later maturity of peanuts is the critical period of aspergillus flavus infection. Intact and strength of peanut's peel and testa plays an important role to aspergillus flavus infection of peanut. The improvement of the composition and the structure for the peal and testa of penut may produce a new kind of penut with high resistence, even immunity to aspergillus flavus. It will have important mean for theory and practice. The best way to reduce the adverse influence of resistence gene on transgenetic plants is to start the resistenc gene expression by specific promoter. It is the best way to construct plant super expression carrier using MAR system in order to prevent silence of transferred genes. The 8,13,L19 and S19 are name of promoter for species expression of peel and testa from our laboratory. TTG2,WRKY60,CBF2 and MYB are transcription factors from our laboratory too.In this research:1. We cloned TTG1 gene, Sequencing result contrasted protein information of genebank with 91% identity.2. Constructing 6 plant expression vectors strengthening construction composition of penut peal and testa with primers(8,L19,S19) and gene(TTG2,MYB61).3. Constructing 5 plant expression vectors enhancing the resistence of penut peal and testa with primers(8,L19,S19) and gene(CBF2,WRKY60,NPR1).4. Constructing 10 plant expression vectors increasing the resistence enzymes synthesis of penut peal and testa with primers(8,13,L19,S19)and gene(RIP,CHI,MCP-1,RS).5. Constructing 2 seed spesific expression vectors with oleosin primers and RS gene.6. Constructing 3 penut peal and testa spesific super expression vectors primers(8,S19)and gene(TTG1,NPR1,RIP). |
