| In order to study the physiology and biochemistry resistance mechanism in grapes to grape phylloxera, the resistance of V.vinifera, V.vinifera-V. labru -sca, rootstocks, HPDs and wild grapes to grape phylloxera (Daktulosphaira vitifoliae Fitch) were evaluated, while, the relationship between the secondary metabolites as well as serval correlative enzymes of grape roots and phylloxer -a resistance were studied. This study will lay a foundation for the further study and provide information for the selection and breeding of varieties or rootstocks. The main results were as follows:1. The resistance levels of the five grape speicies to phylloxera were greatly different. All V. vinifera and V. vinifera-V. labrusca were highly susceptible, on which the total number of eggs phylloxera laied were more than 150 and their tuberosities were more than 30%, belonged to calss 3; Rootstocks including RSB,Gravesac,520A and 225R were immune to phylloxera, on which the development of phylloxera arrested at the very early stage and the phylloxera survival rate were very low. Rootstocks including Fercal,1613C and the HPD were resistant to phylloxera,the tuberosities were lower than 27% and belonged to class 1-2. V.quinquangularis of wide grape was resistant to phylloxera,the tuberosities was 15.45%, belonged to class 2. The other wide grape including V.adenocluda H-M, Henan and a hybrid NW196 were highly susceptible to phylloxera,and they had a tuberosities more than 30%.2. The correlation between grape root phylloxera resistance level and the content of total phenols, flavonoids, lignin and pectin in root phloem was positively significant, whose interrelating coefficient were 0.797, 0.881, 0.885 and 0.919 respectively. In descending order, the contant of the 4 mater in these grape species lead is: rootstocks >HPDs >V.vinifera, V.vinifera-V. labrusca and wide grapes.3. Phylloxera infestation induced secondry metabolism maters and enzyme activity increased in new roots. Resistance level of grape to phylloxera was closely related to the activity of PAL,PPO and SOD, whose interrelating coefficient were 0.965, 0.909 and 0.782 respectively; The PAL, PPO and SOD activity of resistant rootstock 140Ru varied greatly after infected by phylloxera than those of the other three varieties including Tyoho, Tamina and Crimson. The accumulation of total phenols and lignin in infested roots was highly correlative with grape phylloxera resistance, and their interrelating coefficient were 0.987 and 0.841. The content of total phenols and lignin of 140Ru after inoculation was 33.68%, 43.01% and 23.49% higher than that of Tyoho, Tamina and Crimson。... |
