| Gibberellin (GA) is an important plant endogenous hormones and plays a important role in whole plant development such as seed germination, stem and root growth and etc. Ent-kaurene oxidase (KO1) and GA 2-oxidase (GA2ox3) are two key enzymes in GA synthetic pathway. To date KO1 and GA2ox3 genes have not been cloned. In this study, we clone KO1 genes and GA-2ox3 gene in wheat and then transformated their sequences into rice to analyze their potential functions. Meanwhile, we applied the transcriptomic sequencing technology (RNA-seq technique) in two parents of a wheat genetic population, and analyzed the genes in GA synthesis pathway. The main results were as follows:1.14 full-length cDNA sequences of KO1 and GA2ox3 genes were cloned from wheat.2. Transgenic rice lines (To) with over-expressed the two genes were obtained and their phenotypes were observed. No significant phenotype changes were observed in the current lines and further investigation will be done in T1-2 lines.3.1.8 Gb data were generated for each of the two wheat parental cultivars using RNA-seq technique. The data were then assemblyed and annotated. The phylogenetic trees and expression levels of the unique sequences from 7 gene families in GA synthesis pathway were profiled. |
