Rapeseed(Brassica napus L) is one of the main oilseed crops in China. It plays an essential role in the national economy and people's life. Increasing the oil and protein contents is an important way to enhance the use value of rapeseed. Yellow-seeded rapeseed (Brassica napus L) is characterized with thinner seed coat and lower hull percentage, which are correlated with higher oil and protein contents compared to black-seeded rapeseed under the same genetic background. Yellow-seeded coat has been one important breeding goal of rapeseed quality improvement.Aimed at exploring the mechanism of seed coat colour formation in the resynthesized yellow-seeded B. napus line No.2127-17, we constructed the subtractive library of yellow seeds and black seeds, and the main results were as follows:Both subtractive libraries, a forward and a reverse, which were seed color related in resynthesized yellow-seeded Brassica napus line No.2127-17, were constructed using suppression subtractive hybridization (SSH). Then we screened out 60 differentially expressed clones from the subtractive libraries using NILs (NILsl:No.2127-17/94570; NILs2:Yellow sarson/3H129).Sequencing results showed that 60 clones represented 43 different gene fragments. The gene functions mainly involved the following several aspects:lipid and protein storage, energy metabolism, transcription, cellular structure, signal transduction etc.Real-time quantitative PCR analyses of several clones (1F17,2E6,1A14,4E11, 301,4M23), showed that their expressed level in the yellow seeds were higher than that in the black.The clones,1F17,2E6,1A14,4E11 and 301, were related with lipid and protein accumulation. Thus seed oil content was negative correlation with seed coat colour. The clones,4M23 and 2E7, may participate in flavonoids synthesis and affect the seed color formation.
