| Objective: To research the difference and implications of CD59 positive cells,placenta tissue CD59 protein and CD59 gene between the patients with preeclampsia and normal pregnancy women. To investigate the role of CD59 molecules in the pathogenesis of preeclampsia patients.Methods: We selected 57 cases of preeclampsia maternal hospital childbirth. Among of there have 23 pregnant women with severe preeclampsia(preeclampsia group). The other normal late pregnancy were selected as a control group of 30 cases of pregnant and lying in women. The level of CD59 of PBMC were detected by flow cytometry; the protein level of CD59 was detected by immunohistochemistry; the m RNA level of CD59 was detected by real-time quantitative PCR.Results: The FCM research showed that the expression of CD59 was 86.86 + 1.91 in normal pregnancy women, the severe preeclampsia group was 27.26 + 7.05, the expression level of CD59 molecules in severe preeclampsia group was significantly lower than that of in late pregnancy group(P<0.05). After treatment, the number of CD59 positive cell increased in severe preeclampsia group, there was statistically significant difference before or after treatment. The expression level of CD59 in mild and severe preeclampsia placentas group were significantly lower than the normal pregnant women(P<0.05), the difference has statistically significant.Conclusion: The CD59 of cellular level, protein level, gene level in severe preeclampsia group were significantly lower than in normal late pregnancy group.After treatment, the expression level of CD59 was obvious increased in maternal severe preeclampsia group. The results showed that the lower expression of CD59 in PBMC and placenta was closely related to the occurrence of preeclampsia. |
