This is because under normal circumstances, the brain to regulate cardiac activity by the autonomic nervous system,Clinically known as brain-heart syndrome 。 Brain heart syndrome can be a variety of intracranial disorders.IncludeEpilepsy,Acute brain injury,Acute cerebrovascular disease, Intracranial inflammation, Intracranial tumors And cause increased intracranial pressure can cause other diseases,So called brain-derived cardiac damage.Current knowledge, Temporal lobe epilepsy is the most common caused by brain-heart syndrome, Common to the world, an example of multiple epilepsy(In China,Epilepsy annual number of approximately 25.3 / 100,000 population,And the rise),Most of the cases, there may be heart rate, and even cardiac arrest, [1,2] The emergence of a serious risk of death.As such,Brain-heart syndrome research, For the treatment of clinical significance. Calcium-sensing receptor(CaSR) found to make people aware Ca2 + can also be used as extracellular first messenger hormones play a similar role[3].Calcium-sensing receptors belong to the G protein-coupled receptor family member C, In 1993,Brown EM [4]by other first cloned from bovine parathyroid, Subsequent studies confirmed, CaSR is widely present in the thyroid gland, intestine, kidney, liver, stomach, central nervous system and other tissues and organs[5]. Wang et al. 2003 for the first time in the cardiac tissue found CaSR.The present study showed,CaSR extracellular Ca2 + can identify subtle changes, And to ensure that the time regulating intracellular calcium homeostasis, While maintaining the body Ca2 + homeostasis and other metal ions play a key role in homeostasis, And is involved in the regulation of hormone secretion, cell proliferation, differentiation, apoptosis, gene expression, and so the process of ion channel opening[6].In2012, Guo Jin persons confirmed[7],CaSR involved in myocardial ischemia / reperfusion injury, myocardial hypertrophy occurs[8].Epilepsy rat cardiomyocytes CaSR expression pathway and its role has not been reported cases.This study examined the cardiomyocytes pentylenetetrazol induced rat model of chronic epilepsy CaSR expression, Epilepsy rat cardiomyocytes prove there CaSR expression and its relationship with the mitochondria in apoptosis.Objective Establish a chronic epileptic rat model kindled by PTZ and observe the expression of CaSR and mitochondrion pathway of the apoptosis.Methods Adult male Wistar rats were divided into 2 groups randomly.The 12 rats in the normal control group(NC)were injected saline intraperitoneally(i.p) by 3.5ml/kg for consecutive 28 days.The 12 rats in PTZ group received intraperitoneal injection of 1%pentylenetetrazol(PTZ)by 35 mg/kg for consecutive 28 days. The level of plasma creatine kinase(CK),MB isoenzyme of creatine kinase(CK-MB) were measured by ultraviolet spectrophotometry.The myocardial histologic structural changes were observed under the microscope after HE staining.The change of ultrastructure in the myocardium of rats was observed by electron microscopy. EEG tests if it has any abnormal discharge of brain Heart rate and the cardiac function of the rats in each group were detect by echocardiography.The level of CaSR、Bcl-2、Caspase3 and Bax was detected by western blot.Results CK、CK-MB were increased obviously in PTZ group P<0.05.HE staining showed the cell swell. EEG shows epilepsy waves.By echocardiography, rhythm of the heart is increased, the left ventricular wall motion abnormal, and see the rhythmdisorder The change in expression of CaSR、Bax、Caspase3 were increase,while Bcl-2 were decrease p<0.05.Conclusion The level of CaSR expression increased in a chronic epileptic rat model.CaSR activate the expressions of Bax and Caspase-3 of mitochondrion,then influence the cardiac myocyte apoptosis.
