Purpose:Construction:Tibia bone cancer pain in mice model was established, detected of pain behavior, evaluated the analgesic effect of Traditional Chinese medicine analgesia Babu paste, and through the observation groups of mice serum bradykinin,β- endorphin content changes, to investigate analgesic mechanism of the analgesic action of it to bone cancer pain.Material and method:1.Material: C57 BL / 6 mice, 80, 6-7 weeks old, female, body weight 16-18g; β-endorphin mouse ELISA kit, Bradykinin mouse ELISA kit, Lewis lung carcinoma cell line, Micro-injector. Blood cell counter, Olympus BX51 microscope, analgesia Babu paste, low-speed centrifuge, electronic balance, constant temperature water bath, von Frey filaments, Voltaren cream, 6% sodium sulfide solution,D-Hanks solution, DMEM high glucose liquid medium, DMSO, FBS(10% fetal calf serum), 0.25℅ trypsin, double anti-(penicillin- streptomycin), PBS solution,erythromycin ointment, bone wax,EP tube.2.Methods: The mice were divided into eight groups, the normal group, sham operation group, model group, Voltaren group, analgesia Babu paste each dose group(low dose group, the equivalent dose group, middle dose group, high dose group) 10 mouse. Blood samples were taken before surgery, postoperative 7th day,11 st day, 15 th day of mechanical paw withdrawal response was measured. The 15 th days, after measuring pain threshold, each group of mice eyeballs blood was collected,enzyme-linked immunosorbent assay(ELISA method) serum bradykininβ-endorphin peptide content was tested.The result:.1. Paw withdrawal response in mice pain threshold experiments showed that: Before modeling, model group mice pain threshold compared with normal group, sham group,(P> 0.05) was not statistically significant. 7th,11 st,15th days after the first pain threshold were measured in each group of mice, 7th days,the first 11 days,the model group, analgesia Babu paste each dose group, Voltaren group pain threshold began to decrease, with the normal group, sham group have more significant difference,(P <0.05) was statistically significant, the 15 th day(treatment day 5), normal group and model group, analgesic paste each dose group,Voltaren group,(P <0.05) pain threshold was statistically significant.Equivalent dose group, middle dose group, high dose group, Voltaren pain threshold compared with the model group, low-dose group were significantly increased,(P <0.05) was statistically significant, Comparison between the low-dose group and model group,(P> 0.05) was not statistically significant,Middle dose group, high dose group compared with Voltaren group,pain threshold(P> 0.05) was not statistically significant.2.Measured by ELISA assay in serum levels of mice in each group β- endorphin(β-EP) content. The experimental results showed that: Modeling of bone cancer pain mice in each group content of β-EP and the normal control group, sham group increased significantly(p <0.01) considered statistically significant.Traditional Chinese medicine analgesia Babu paste, the high-dose group, Voltaren with model group, low dose group, the equivalent dose group had a significant increase, p <0.01 considered statistically significant.3.Measured By ELISA assay in serum levels of mice in each group bradykinin(BK)content. The experimental results showed that: Bradykinin content modeling of bone cancer pain mice in each group and the normal control group, sham group increased significantly, p <0.01 considered statistically significant.Traditional chinese medicine analgesia Babu paste,the high-dose group of,Voltaren with model group, low dose group, the equivalent dose group had significantly reduced, p <0.01 considered statistically significant.Conclusion:1.Traditional Chinese medicine analgesia Babu paste can increase the pain threshold of bone cancer pain mice, Its analgesic effect is close to Voltaren,Analgesic effect of high-dose group than in the equivalent low-dose group.2.Traditional Chinese medicine analgesia Babu paste can increase bone cancer pain model in mouse serum β-endorphin.3.Traditional Chinese medicine analgesia Babu paste can reduce the amount of bone cancer pain modelin mouse serum bradykinin. |
