Comparative Study On The Effect Of Electroacupuncture At "four - Off" Group And Corresponding Single - Point On Brain Monoamine Neurotransmitters In PSD Model Rats

Objective:To observe the effect of electroacupuncture(EA) of "Si-guan" points[bilateral"Hegu"(LI4)-"Taichong"(LR3)] and corresponding single point on ethology and cerebral monoamine neurotransmitters in Poststroke-depression(PSD) Rats; to investigate the synergistic effect and the possible mechanism of bilateral LI4-LR3.Methods:Sprague-Dawley rats were quadratic randomly divided into five groups (8rats/group):control, model, EA group-1("Si-guan" points), EA group-2(bilateral LI4points), EAgroup-3(bilateral LR3points). The PSD model was established by separately breeding and chronic unpredictable moderate stress (CUMS) after middle cerebral artery occlusion (MACO).EA(30min) LI4-LR3, bilateral LI4and bilateral LR3were applied in group-1、EA group-2and EAgroup-3respectively, once daily, continuously for21days. The indexes of the behaviors of PSD model rats were measured by the methods of Open-Field test and Sucrose solution consumption test,the fluorometer detecting system of HPLC was used to mensurate the monoamine neurotransmitters of Rat Hypothalamus.Results:1.After operration,Compared with control group,the data of SD rats eating and inflow、body mass growth index、behavioral science and brain monoamine neurotransmitter levels were significantly reduced (P<0.01);2.After treatment,Compared with the model group,the eating and infiow、body mass growth index、nevrous functional defects、radio of the consumption of sugar waterlevels of EA group-1("Si-guan" points), EA group-2(bilateral LI4points), EAgroup-3(bilateral LR3points) all improve obviously (P<0.01);3.After treatment,Compared between EAgroup-1,EAgroup-2and EAgroup-3,EAgroup-1in improving the body quality growth effect is best;EAgroup-2in the aspect of improving sugar water consumption rate effect is best;EAgroup-3In improving open-field test number of upright, increasing the prefrontal cortex NE content and hypothalamus area5-HT and DA content effect is best.Conclusion:Compared with control group,By separately breeding and chronic unpredictable moderate stress after middle cerebral artery occlusion for preparation of Post-stroke depression Model,Rats in food intake, body quality rise slowly, behavioural indicators test results is reduced, in the prefrontal cortex, hippocampus, hypothalamus area of the rats’ brain monoamine neurotransmitter content in different degree; Electroacupuncture both of EA "Si-guan" points and bilateral "Taichong" points can significant improved Eating and inflow reduce, body quality rise slowly, determination of neural function and reduce behavioral test result of PSD rats, the mechanism neurotransmitters perhaps related to the increase of monoamine in Rats’brain。...
