| Purpose:The bile reflux gastritis animal model was prepared by feeding rats refluxliquid. The rats were cured by Jiangni Hewei decoction. The influence of theJiangni Hewei decoction on serum interleukin-6(LI-6), tumor necrosis factor(TNF-a), gastrin (GAS), endothelin (ET) levels, changes of gastric mucosalpathology of animal model were examined and compared with the HydrotalciteTablets. The results were used to clarify the mechanism of the effect of JiangniHewei decoction on gastric mucosa injury of the rat bile reflux gastritis.Materials and experimental procedure:1. Grouping model and interventionIn order to adapt the laboratory environment, the selected72healthy maleWister rats which will be used in the experiments were fed for3days. And then,according to the weight of rats, the rats which arrives requirement were divided6groups. They are the blank group, model group, the model group of Jiangni Heweidecoction with high dose, the model group of Jiangni Hewei decoction with middledose, the model group of Jiangni Hewei decoction with low dose, the comparinggroup of positive medicine control, gender each half. Except the blank groupfed equal volume of distilled water, another5groups of rats were fed the refluxliquid prepared by ourselves for15days. The self-made reflux liquid was preparedwith2.5g sodium cholate,1.5g pancreatin,0.25g lecithin (abxing) and100mldistilled water. The rats were fed the1.5ml/l00g reflux liquid two times everyday. After the model established, the model group and blank group also were fedthe equal volume normal saline and the positive medicine control group wastreated with Hydrotalcite Tablets. The doses of the traditional Chinese medicinewith high, middle and low dose group were37.2g,18.6,9.3/kg body weight,respectively.2.The observation of the general conditionThe mental state, diet, situation after twice defecates, the body weight after filling gastric by reflux liquid, hair color and death etc. of rats werecarefullyrecorded during experiment.3.Experimental indexes and detectionAfter gavage for30days, the rats were taken. The rats were fasting and waterdeprivation for24hours after the last gavage. At the same day as the rats taken,the rats were weighed and then the anesthesia was injected according to the bodyweight. The2ml blood was obtained from the abdominal aortic to test the levelof serum LI-6, TNF-a, GAS and ET-1.The stomach was cut under aseptic conditionand then cut open along the big bend. The stomach contents were washed with icesaline. It was dried by filter paper and then was placed into the polyformaldehyde solution fixation. Finally, the pathological changes of gastricmucosa were observed under light microscope.Results:Part1: the influence of Jiangni Hewei decoction on serum LI-6, TNF-a, GASand ET-2content of the model rats of bile reflux gastritis.The content of serum IL-6, GAS, TNF-a and ET of different groups wascompered.Compared with the model group, the Jiangni Hewei decoction groups withhigh, medium and low dose and the positive medicine control group could increasethe content of IL-6, GAS, and TNF-a To reduce ET, with statistical significance(P<0.05). Compared with the positive medicine control group, the IL-6, TNF-a,GAS and ET of the Jiangni Hewei decoction with high, medium and low dose groupshas no obvious change and there is no significant difference (P>0.05).Part2: the influence of Jiangni Hewei decoction on bile reflux gastritisgastric mucosa pathological model of ratsThe gastric mucosa of the blank group rats is smooth, detachment and undamaged.The inflammatory cells are rarely seen. The gastric mucosa of rats in the modelgroup shows separation and damage. A large amount of inflammatory cells wereobserved. Compared with the model group, the detachment and damage of gastricmucosal cells of the Jiangni Hewei decoction groups with high, medium and low dose and the positive medicine control group is lighter and the infiltrationof inflammatory cells is less. The high dose group of the Jiangni Heweidecoction has the best effect, followed by the medium dose and positivemedicine control group. Compared with the model group, the results of semiquantitative index of inflammatory cells showes that the inflammatory cellsof the infiltration alleviate of the Jiangni Hewei decoction of the high doesgroup, the low dose group, positive control group and Western medicine waslighter (P<0.05).Conclusions:1In curing the bile reflux gastritis, the Jiangni Hewei decoction has goodeffectiveness. It obviously increases the level of interleukin-6, tumor necrosisfactor-a, gastrin and endothelin of the bile reflux gastritis rats and enhancesthe protection of gastric to reduce gastric mucosal injury.2In curing the bile reflux gastritis, the Jiangni Hewei decoction can reducethe infiltration of inflammatory cells, To strengthen the protection of gastricmucosa, reduce the damage of gastric mucosa. |
