| Objective:To study the protective effects on acute liver injury induced by D-galactosamine in rat, the test samples include the water and methanol extract of Cortex Acanthopanacis, the water and methanol extract of Acanthopanax gracilistylus Leaf, the total glycosides extract, the total genin extract, and impressic acid which are both derived from Acanthopanax gracilistylus Leaf. Research the acute toxicity experiment of effective samples. To establish the HPLC analysis method, carry out the content determination of main components.Methods:Using the water decoction and methanol extraction method, the preparation of drug are as follows:Cortex Acanthopanacis water extract, Cortex Acanthopanacis methanol extract, Acanthopanax gracilistylus Leaf water extract, Acanthopanax gracilistylus Leaf methanol extract; the total glycosides extract, total genin extract and impressic aicd were derived from Acanthopanax gracilistylus Leaf. Rats as the research object, based on the D-galactosamine acute liver injury mode in blood setum glutamic-pyruvic transaminase (ALT), glutamic-oxalacetic transamineasa (AST), total bilirubin (TBIL), superoxide dismutase (SOD), malondialdehyde (MDA), nitric oxide (NO), as observation indexes, observe the effects of test substances on animal model. The acute toxicity experiment was research to the samples which have a protective effect to acute liver injury rat. HPLC method was used for content determination of the main components in effective samples to control its quality.Results:â‘ Cortex Acanthopanacis medicinal materials150g, obtain water extract350ml; Cortex Acanthopanacis medicinal materials150g, obtain methanol extract12.3g; Acanthopanax gracilistylus dried Leaves150g, obtain water extract350ml; Acanthopanax gracilistylus dried Leaves150g, obtain methanol extract39.3g.â‘¡The content of serum ALT, AST, TBIL, SOD, MDA, NO was significantly different between model group and blank group. Results of pathological section showed that there is serious liver injury in the rat of model group. The positive drug group which diammonium glycyrrhizinate was intervened showed a much lighter liver injury compared with model group. The group of total genin extract of Acanthopanax gracilistylus Leaf (100mg/kg) also showed a protective effect to liver injury, the effect is almost equivalent to diammonium glycyrrhizinate. â‘¢The acute toxicity experiment of total genin extract of Acanthopanax gracilistylus leaf proved that the extract is safe. When50.1times of effective dose was given to the mouse of treatment group, no abnormal situation and no death, no obvious difference between the weights of groups.â‘£The method is accurate, simple, and can be used as assay of3a,11a,23-trihydroxy-lup-20(29)-en-28-oic acid,3a,lla-dihydroxy-23-oxo-lup-20(29)-en-28-oic acid, impressic acid and acankoreanogein in leaf total glucoside extract of Acanthopanax gracilistylus W.W.Smith.Conclusion:The total genin extract of Acanthopanax gracilistylus Leaf can inhibit the decrease of serum ALTã€ASTã€TBILã€MDA and NO of D-GaIN induced acute liver injury rat, which also inhibit the increase of serum SOD.The total genin extract alleviate necrosis and degeneration of hepatocyte. |
