Objective: Human cytomegalovirus(HCMV) is one of the largest virus of human herpes virus. Human is the only host of HCMV and most of HCMV-Infected people is asymptomatic., The virus can be activated and result in serious infection when the HCMV-Infected people is in the low immunity(such as organ transplantation, AIDS and so on). HCMV infection is all over the world and the infection rate is very high. Because the virus is difficult to be removed completely from the body and the cost of treatment is very expensive, the establishment of the early laboratory diagnosis about HCMV infection is very important to prevent and cure diseases. we used i TRAQ to do the identification and quantitative analysis of serum protein on the HCMV-infected people,which is in order to find proteome expression profiling between HCMV-Infected people and normal people,and to identify the HCMV protein-biomarker.This is not only for conducting the quick and accurate effectiveness evaluation of the treatment in the early diagnosis for the future study,but also provide scientific experimental basis for the future research and developing the rapid and specific serological diagnostic reagent.Methods: Ten cases of HCMV infection serum and Ten cases of normal serum was collected to extract serum protein and remove high abundance proteins and obtain peptides by tryptic digestion of protein after acquiring.Appling the analytical method based on eight-plex isobaric tags for relative and absolute quantification coupled with multiple chromatographic fractionation and tandem mass spectrometry to mark between HCMV-Infected people and normal people.Using the Separation of liquid chromatogram and analysis of mass spectra and level-2 mass spectrum database to identify the protein and compare the difference of expression of the protein. We found the proteins that have a higher correlation with HCMV infection by searching experimentally-derived protein as keywords in NCBI Search engine and literaturereviewing and analysing.Results: We used the total protein in human serum as the research object and conducted protein identification and proteomic analysis by comparing serum protein between HCMV-Infected people and normal people to protein identification,a total of 362 proteins were identified.We also found that the expression of 49 different kinds of proteins up-regulated more than two times,in which 19 kinds of preteins for increase and 30 kinds of preteins for reduction.And found that Adiponectin,Alpha-1-antitrypsin,Apolipoprotein E,C-reactive protein,Haptoglobin and Serum amyloid A-1 protein have a higher correlation with HCMV infection by using NCBI Search engine.Conclusion: We got the Serum proteomic differential expression patterns between HCMV-infected people and normal people by using quantitative proteomics technology. It is concluded that C-reactive protein and serum amyloid protein, adiponectin and apolipoprotein A1 this four kinds of protein related to HCMV infection as well as atherosclerosis; Alpha-1 antitrypsin and haptoglobin related to HCMV infection as well as occurrence of cancer, so this six kinds of proteins can be identified preliminarily as protein-biomarker of HCMV infection,which provided scientific experimental basis for the research on the pathogenic mechanism of HCMV infection.
