This paper includes two parts Reviews and Experimental Research.Part One:ReviewsThe expression of SAMP8 AD mice and histological observation and ADAM 10 and ADMA17 in cerebral cortex of mice, to investigate the effect and mechanism of electroacupuncture intervention in AD, and compared the difference between electroacupuncture and drug treatment of AD. 1:Methods:This study used rapid aging (SAMP8) mice as a AD model, the core part of the amyloid precursor protein and the pathogenesis of AD (amyloid precursor protein, APP) related to metabolism ADAM 10 and ADAM 17 as the starting point, the changes of mice were observed after treatment, also intends to use immunohistochemistry and Western blot the techniques and methods to study the effect of Electroacupuncture on AD mice cerebral cortex ADAM 10 and ADAM 17, and its mechanism, and compared the difference between electroacupuncture and drug treatment of AD.2.Results:1) behavioral results:The navigation experiment results:1) mice at different time points between groups comparison results showed that:with the C group (normal control group), M group (model group) 5 days of escape latency were increased in fifth days was statistically significant difference between the groups (P<0.05); compared with C group, D group (the medicine group) 5 days of the escape latency was shortened, but the difference was not statistically significant (P>0.05); compared with C group, AE group (EA group) for 5 days the escape latency was shortened to fifth days, was statistically significant difference between the groups (P<0.05); compared with D group, PE group, escape latency than in the D group, but no significant difference in efficacy between the two (P>0.05).2) mice at different time points within group comparisons showed that:compared with first days, C group and D group, AE group in fifth days of training when the escape latency was significantly reduced, and the difference was significant (P<0.05); M group in the process of training, the escape latency changes had no significant difference (P>0.05).3) four groups of mice escape latency decreased trend. The C group showed a downhill downward trend; D group fourth days to start the escape latency appeared downhill decline; AE group third days to start the escape latency appeared downhill decline; M group decreased slowly, the rise in third days, the latency trend is random.Space exploration results:compared with B group, M group significantly shorten the distance in the target quadrant, there was significant difference (P<0.01); compared with C group, D group and distance in the target quadrant was significantly increased, significant difference (P<0.05); compared with C group, AE group and distance in the target quadrant was significantly increased, and there is a significant difference (P<0.01). Compared with D group, AE group and distance in the target quadrant increased, but no significant difference (P>0.05)2) the results of immunohistochemistry:IOD through the statistical analysis on gray value of positive cells, compared with C group, B group, D group, PE group, ADAM 10 expression in the cerebral cortex of AD mice increased, and the differences were significant difference (P<0.01); compared with the D group, after acupuncture intervention, Electroacupuncture group in ADAM 10 AD model mice cerebral cortex increased slightly, but the difference was not statistically significant (P>0.05).3) Western blot results:the value of statistical analysis by gray bands, compared with C group, B group, D group, PE group, ADAM17 expression in the cerebral cortex of AD mice increased, and the differences were significant difference (P<0.01); compared with the D group, after acupuncture intervention, electroacupuncture group ADAM 17 in AD model the mouse cerebral cortex increased slightly, but the difference was not statistically significant (P>0.05).3.Conclusion:1) the results showed that:M group than in C group of mice had obvious impairment of learning and memory, that SAMP8 mice have similar symptoms of AD, is a quasi AD model;2) drugs and acupuncture intervention of AD mice, improve the ability of learning and memory were, there is no difference between the two effects;3) drugs and acupuncture intervention of AD mice, the expression of ADAM 10 and ADMA17 were increased in the cerebral cortex of SAMP8 mice, which play a role in treatment of dementia, and the therapeutic effect is better than drugs;4) the mechanism of donepezil hydrochloride in the treatment of AD is to inhibit the degradation of acetylcholine, this experiment through drug intervention ADAM10 and increased ADMA17 were expressed in the cerebral cortex of SAMP8 mice, donepezil may also consider the effect on the alpha secretase degradation pathway for the treatment of AD;... |
