| ObjectivesThis study based on the replenishing Qi resolving stasis and removing toxin compound formula,which belongs to the national TCM expert Professor Du Huaitang of Dongzhimen Hospital of Beijing University of Chinese Medicine.Through designing the prevention test and the treatment test,the rats with acute lung injuries/acute respiratory distress syndrome (ALI/ARDS) due to lipopolysaccharide(LPS)were studied to explore the protective effect of replenishing Qi resolving stasis and removing toxin compound formula on the rats with ALI due to LPS,and to observe the effect of the serum IL-8 and TNF-a levels.MethodsThe Prevention Test:32 healthy male SD rats were randomly divided into 4 groups (n=8).According to the animal husbandry standards,all the rats were fed for one week in advance.The ALI model was prepared with LPS intraperitoneal injection by 5mg/kg.The low-dose Chinese medicine group and the high-dose Chinese medicine group were modeled with LPS(5mg/kg) after were given low-dose Chinese medicine or high-dose Chinese medicine for 14 days. Rats in the model group while rats in the blank control group were with no process after normal diet for 14 days. The rats of the 4 groups were executed with anesthesia after 6 hours,and then immediately collecting abdominal aortic blood and lung tissue.Serum specimen and lung tissue HE staining biopsy were produced and the contents of IL-8 and TNF-a in serum were measured with enzyme-linked immunosorbent assay.The Treatment Test:24 healthy male SD rats were randomly divided into 3 groups(n=8). According to the animal husbandry standards,all the rats were fed for one week in advance. The ALI model was prepared with LPS intraperitoneal injection by 5mg/kg.Making the better one between the low-dose Chinese medicine group and the high-dose Chinese medicine group of Prevention Test as the Chinese medicine group in Treatment Test.The Chinese medicine group were immediately given Chinese medicine after modeled with LPS(5mg/kg).Rats in the model group were with no intervention after LPS modeling,and rats in the blank control group were with no intervention.The rats of the 4 groups were executed with anesthesia after 6 hours,and then immediately collecting abdominal aortic blood and lung tissue.Serum specimen and lung tissue HE staining biopsy were produced and the contents of IL-8 and TNF-a in serum were measured with enzyme-linked immunosorbent assay.ResultsFor visual observation,the lung surface of rats in blank control group was ruddy, and there was no light red fluid effusion from the section.Rats in model group appeared a significant lung tissue edema and the lung surface became dark red, while there was light red fluid effusion from the section.The lung surface of rats in low-dose Chinese medicine group was ruddy, and there was a little light red fluid effusion from the section.The lung surface of rats in high-dose Chinese medicine group was light red,and there was a little light red fluid effusion from the section.For pathological observation,the alveolar structure was complete and clear in blank control group, and there was no edema for alveolar septum, edema fluid in the alveolar lumen, and inflammatory cell infiltration in lung tissues. In model group, the Structure of rat were damaged obviously,and it appeared alveolar and pulmonary interstitial edema,lots of inflammatory cell infiltrations,alveolar wall thickening,alveolar lumen narrowed and distorted, part of alveolar collapsed,lots of secretions containing red blood cells in bronchial lumen and alveolar lumen,and the pulmonary capillary congestion and edema. Rats of the low-dose Chinese medicine group were lighter than those of the model group.In the high-dose Chinese medicine group,the alveolar structure was complete.And the bleeding, exudation,microvascular edema were reduced.About serum IL-8,TNF-a levels,the model group was higher than the blank control group(P<0.05),and the high-dose Chinese medicine group and the low-dose Chinese medicine group both decreased than the model group,while rats in the high-dose Chinese medicine group were lower than the low-dose Chinese medicine group(P<0.05).Treatment test:For visual observation,Rats in blank control group appeared no lung tissue edema.The lung surface was ruddy,and there was no light red fluid effusion from the section.The lung surface of rats in model group became dark red, and there was a little light red fluid effusion from the section. While The lung surface of rats in Chinese medicine group was light red.For pathological observation,the alveolar structure was complete and clear in blank control group, and there was no edema for alveolar septum, edema fluid in the alveolar lumen, and inflammatory cell infiltration in lung tissues.The lung structure of the model groud were damaged obviously,and there was inflammatory cell infiltration in lung tissues.the lung tissue injury in rats of Chinese medicine group were reduced compared with the model group.About serum IL-8 and TNF-a level,the model group was significantly higher than the blank control group(P<0.05). While the Chinese medicine group was significantly higher than the model group(P<0.05).ConclusionsReplenishing Qi resolving stasis and removing toxin compound formula can reduce the lung tissue injuries of rats with LPS-induced ALI,and relieve the level of congestion,edema and inflammatory cell infiltration in bronchial lumen and alveolar lumen.The protective effect of replenishing Qi resolving stasis and removing toxin compound formula may be related to reduce rats’serum IL-8 and TNF-a level,reduce inflammation reactions.through this sutdy,we found that the frequency spectrum water had anti-inflammatory effect,and frequency spectrum water with Chinese traditional medicine compound could improve the effect.By changing the conventional solvent,Chinese traditional medicine can realize synergistic effect. |
