| Objective:Study to investigate the efficacy of viola for the treatment of inflammatory diseases.Methods:Using viola water decoction lavage intervention in clean grade SD rats of conventional preparation containing containing serum, In a certain concentration coculture with mouse peritoneal macrophages in96-well plates. After24hours, NO content were determined in the culture supernatant by Griess colorimetry, cell phagocytosis activity was determined by neutral red uptake assay, TNF-a and IL-6content were determined in the culture supernatant by ELISA, changes in expression levels of TLR-1ã€TLR-2ã€TLR-3ã€TLR-4ã€TLR-5mRNAs were determined by RT-PCR.Results:Compared with the same concentration of normal serum group,5%-20%of the role of viola-containing serum:Without LPS activated macrophages increase secretion of TNF-αã€NO (P<0.05or P<0.01). Enhanced expression of iNOS (20%containing serum group)(P<0.01).And decrease secretion of IL-6(5%containing serum group)(P<0.01) or increase secretion of IL-6(10%containing serum group)(P<0.01); Macrophage activity is significantly increased (P<0.01); Within certain concentration, viola containing serum group with increased or decreased expression levels of TLR-1ã€TLR-2ã€TLR-3mRNA (P<0.05or P<0.01), the expression level of TLR-4was not significantly altered in each group (P>0.05), and the expression level of TLR-5was significantly increased in each group (P<0.01)(2) With LPS activated macrophages decrease secretion of TNF-αã€NO (P<0.05or P<0.01), Inhibit expression of iNOS (5%~10%containing serum group)(P<0.01), And decrease secretion of IL-6(10%containing serum group)(P<0.01) or increase secretion of IL-6(20%containing serum group)(P<0.01); Compared with the same concentration of without LPS activated macrophages group, Normal serum5%~20%with various concentrations of LPS-activated macrophage TNF- a, IL-6, NO increased secretion and promote iNOS expression (P<0.05or P<0.01);(2) Viola-containing serum5%~20%with various concentrations of LPS-activated macrophages significantly decreased secretion of NO〠TNF-α in5%containing serum group (P<0.01), increase secretion of TNF-a in10%and20%containing serum group (P<0.01); Enhanced expression of iNOS in5%containing serum group (P<0.01), But inhibit expression of iNOS in10%and20%containing serum group (P<0.01); decrease secretion of IL-6in5%containing serum group (P<0.01), And increase secretion of IL-6in20%containing serum group (P<0.01)Conclusions:1. Intervention effect of viola-containing Serum on macrophages to produce TNF-αã€IL-6ã€NOå’ŒiNOS which may be the mechanism how it regulation of the inflammatory response;2. Viola-containing serum has obvious promoting effect on macrophage phagocytosis, the mechanism of which may be related to that this drug regulates and controls part of macrophage TOLL-like receptor expression. |
