ObjiectiveTo observe the effects of The mesenchymal stem cells(MSCs) transplantation combined with traditional Chinese medicine (BU NAO I HAO) on the cognitive function and spatial orientation in vascular dementiarats.MethodsSD rats, which were picked out by water maze test, were randomly divided into Sham-operated, model, transplant, transplantion with Chinese herb-compoud groups. All rats inducted by D-galactos(D-gal) were treated with ALCL3for6weeks.7days after rat model of VD was established, we grafted mesenchymal stem cells into the hippocampus of the rat in transplant, transplantion with Chinese herb-compoud groups.2hours later, rats in transplantion with Chinese herb-compoud group were fed with herb(BU NAO I HAO)for4weeks,2times a day. After the treatment, we detected the cognitive function and spatial orientation, also the activity of SOD and content of MDA, Ach in cerebral tissues were determined.End we assayed the SYP protein expression with Western blotting.ResultsThe latencies、the swinmming distances and the times across platfof water maze test in model group were significantly longer than those in sham-operated group(P<0.01);and there were significant di fference in the activity of SOD, the content of MDA, Ach and the AchE activity detection.Also there were significant difference in SYP protein expression (P<0.01), Compared with model group, the latency and the swimming distances was shorter and the frequency of crossing platform increased(P<0.01-P<0.05), which were of statistical significance, in transplants group and trandplantion with Chinese herb-compoud group. In ransplantion with Chinese herb-compoud group,the content of MDA were lover and the content of SOD were increased, compared with the model group, there were significant difference(P<0.01). As to the Sham-operated group, there were no significant difference(P>0.05). In transplant group, the activity of SOD were increased. Compared with the model group, there were significant difference (P<0.01). As to the Sham-operated group, there were no significant difference(P>0.05). Both in transplant group and transplantion with Chinese herb-compoud group, the content of Ach and the expression of SYP protein were increased. AchE activity and Ach content detection results instead. Compared with the model group, the difference in transplant group(P<0.01)and ransplantion with Chinese herb-compoud groups (P<0.05)were significant.ConclusionsThe mothed is suit for rats with Alzheimer, s Disease. Compared with the modle group, the ability of learning and memory were improved in transplans group, but in transplantion with Chinese herb-copoud group the improvement were more significant. MSCs transplantation combining BU NAO I HAO therapy can improve the cognitive funvtion and spatial orientation, improving free radical metabolism, enhancing the antioxidant abilitie, reduce the activity of acetylcholine transferase, repairing the injury of SYP.
