Object Preparing nano yam polysaccharide colon-specific capsule, and its quality, in vitrorelease and stability need to be examined. Study of the antioxidant activity and anti-agingeffects of nano yam polysaccharide colon-specific capsule on aging model wistar mice by injectD-galactose in vivo.Methods The ball milling method was used for the preparation of yam polysaccharidenanoparticles; laser particle size distribution instrument was used to analyze the particle sizedistribution; SEM was used to observe the surface morphology; preparing nano yampolysaccharide colon-specific capsule; phenol-sulfuric acid method was used to determine thecontent of polysaccharide in preparation; quality analysis of nano yam polysaccharide colon-specific capsule. Studies on antioxidant indexes superoxide anion as free radical and hydrogenperoxide, hydroxyl radicals, DPPH, reduction potentials and metal chelating in vitro. Todetermine whether the nano yam polysaccharide colon-specific capsule has antioxidant activityand anti-aging effects in vivo, we measured the changes in mice of MDA, SOD, CAT, GPx,LPO, POD, ROS and Na+/K+-ATP enzyme activity assay by using the method of chemicalreagent kit, take Wistar mice into six groups after subcutaneous injection at back of the neckwith D-galactose for40days.Results Yam polysaccharide nanoparticles prepared by high energy ball milling isspherical,its surface is round and even, no adhesion,80%of its particle size concentrated in the575nm;nano yam polysaccharide colon-specific capsule meets the capsule quality requirementsof Chinese Pharmacopoeia2010version. The stability test results show that this product isunstable to heat, hence the storage conditions of nano yam polysaccharide colon-specificcapsule should be “sealed, cool and dry place to save”. In vitro antioxidant experiments showthat, hydrogen peroxide, and metal chelating agent of free radical DPPH, reduction potentialclearance into growth trend with the increase of the concentration of polysaccharide, superoxygen anion free radical and hydroxyl free radical oxidation resistance also and polysaccharideis closely relative to the concentration. We found that the serum and the MDA level of organweights (lung, heart, liver, spleen and kidney) in mice significantly change compared with thosefrom the normal control group (P<0.05). The serum and the organs levels in the experimental group significantly increased in a dose-dependent manner when the nano yam polysaccharidecolon-specific capsule concentration in the SOD, CAT, GPx, LPO, POD, ROS and Na+/K+-ATP activity, while the high dose group was obviously (P<0.05).Conclusion The technology of preparing nano yam polysaccharide colon-specific capsuleis reasonable and practicable; the quality meets the capsule quality requirements of ChinesePharmacopoeia2010version. D-galactose can be successfully used to create aging model wistarmice. These experiments demonstrate that the functional dosage form, prebiotics nano yampolysaccharide colon-specific capsule, exhibits antioxidant effects in vivo with increasing doselevels, especially in the high dose group. These results are valuable for future against oxidativestress to a certain extent use.
