Circulating Immune Complexes Of Several Sorts Of Poultry

The experiment investigated the circulating immune complexes of healthy duck,chicken, black-bone chicken and pigeon.The serum of poultry was deposited several times by PEG precipitation method,and then analyzed by SDS-PAGE.The results reveal after using PEG precipitation method one time,we can receive the real circulating immune complexes.The content of CIC in the four poultry were determined by spectropHotometer.The CIC were dissolved in 0..lmol/L NaOH,then measurated the absorbency after 450nm in spectrophotometer.From the standard curve of human's serum,we can calculate the content of CIC in the four poultry.The results reveal whether the concentration of PEG is 2% or 3.5%,the content of CIC is duck>chicken>black-bone chicken>pigeon.There are significantly differences in levels between the two PEG treatment of each poultry.There are also significantly differences in levels among the four sorts of poultry in the same concentration of PEG.In SDS-PAGE ,we used 10% separation gel and 4% stacking gel. And in SDS-PAGE we used protein Marker analyzing the CIC in healthy poultry.From the electropherogram,we can see chicken and black-bone chicken have the bands of heavey chain and light chain in IgG and IgM.But duck and pigeon only have one antibody.We used Dot-ELISA proving what the antibody is. There are some differences among the four poultry in the band of electropherogram.And the differences between chicken and black-bone chicken which belong to Galliformes are not very obvious in the band of electropherogram.Furthermore,we use Dot-ELISA analyzed the CIC of the four poultry. Determined the conditions of Dot-ELISA .The proper coating time is 60min, The concentration of the Rabbit anti-Chicken IgG-HRP is l:500.The titer of chicken we can measured is 1:1024,and the titer of black-bone chicken is 1:256.From the results we can see the PEG precipitate can reaction with the Rabbit anti-Chicken IgG-HRP.The CIC of the four poultry can isolate by 2%PEG and 3.5%PEG.And we also proved the antibody in the CIC of duck and pigeon is IgG.The CIC of healthy duck,chicken, black-bone chicken and pigeon were isolated by 4%PEG and 3.5%PEGThe CIC were characterized by SDS-PAGE and Dot-ELISA.We also determined the content of CIC in the four poultry by spectrophotometer. Form these results,we hope we can discussed the diagnose and therapy of poultry's disease from the angle of immunology.
