| Peanut (Arachis hypogaea L.) is an important oil and economic crop. Disease, insect and pest grass are affecting the productivity and quality of peanuts. The papers studied on peanut plant regeneration system by somatic embryogenesis and transformation of Rs-afp1 gene by agrobacterium- mediated, tried to gain a peanut anti-fungal disease and improve the productivity and quality of peanuts.1.Optimize of plant regeneration system by somatic embryogenesis in peanutStudied on the effect of genotype and concentration of 2,4-D on plant regeneration by somatic embryogenesis which were used three varieties (Fenghua2, Luhua11, Nongda818) of peanut as source of explants.Then systematic studies were done on the effect of induction days and hormone on plant regeneration from embryonic leaflets of Fenghua2. Study results showed that induction efficiency of MSB+15mg/L 2,4-D was the best and the regeneration capability of Fenghua2 was highest among the three varieties. Fenghua2 was an ideal variety for plant regeneration by somatic embryogenesis, its plantlets rate of callus was 34%. Induction days had an effect on the re-differentiation of callus, study showed that the best induction days was 21d. The callus kept in de-differentiation cultured in medium adding 30mg/L 2,4-D, if they were transfered to base medium could regenerate plantlets, its plantlet rate was 47.4%. Different kinds of hormones had different effects on the re-differentiation of callus, study showed that the best re-differentiation medium was MSB+5mg/L BA. Picloram induced embryonic leaflets to de-differentiation was propitious, but it was bad to induce re-differentiation, and the callus couldn't form plantlet.2.Study on influencing factors of peanut genetic transformation mediated... |
