Sinorhizobium fredii is a fast-growing Chinese isolate with fewer extracelluar polysaccher that has been adopted as a model organism for the study of nitrogen fixation and suited for commercial inoculation production . In previous work, A mutant strain GXHN100 was screened from Tn5gusA5 random insertional mutants of S.frediis train HN01. Plant test showed that GXHN100 nodulated later on soybean and its nodulation efficiency and competitiveness were decreased. Sequencing analysis showed that an open reading frame, named pmrA, was inserted by the Tn5gusA5. A positive clone, named pGXHN300 which containing pmrA fragment of S.fredii strain HN01, was isolated from gene library of S.fredii HNOl by colony in situ hybridization . The aim of this work is to identity novel genes associative nodulation competition.The flanking sequence 8.0kb of prmA was obtained by subcloned and scpuenced.
