| 7 strains of APP from delta district of ZhuJiang was isolated and the serotype was determined, the experiments of culture character, satellitism, CAMP test, sugar ferment test and urease test was accord of APP biology feature, 442bp DNA fragment of APP APX IVA was amplified by PCR, the animal test shows that the 4 strains of APP were pathogenic, sero-typing test shows that 3 strains were serotype 1I, 2 strains were serotype 7, one was serotype 3 and the other was serotype 4. IHA test was proceeded in 31 intensive hogpens of delta district of ZhuJiang, 450 serum samples have been tested, the result shows that the positive rate of APP antibody was 58.4%, and, the serotyping shows that the main prevented APP serotype is 1, secondly serotype 7 and 3.The best nutrition and culture condition of the production of inactivated vaccine was developed, using 2 high virulence APP strains as the strains for inactivated vaccine. The concentration of the bacteria grow in the optimized condition can reach 2.4 billion/ml. Safety test and checking of physics and chemistry feature demonstrated that the vaccine safe and effective. Rabbit and swine were immune, the ebb and flow rule shows that oil emulsion adjuvant inactivated vaccine have better immunogenicity than activated alumina adjuvant vaccine, proceed the secondly immune in the right time can boost the antibody level significantly and have a longer protected period. The immunity effect test shows that swine can be prevent infection from the two serotype of APP by using the bivalent vaccine. Field experiment of the vaccine shows that the swine have a lower feed : meat rate, higher market weight, shorter age come into market, lower mortality and illness, all of above indicate that the bivalent vaccine for local area can be popularized in clinic. |
