| Objective: The Blood Stagnation Syndrome model was made in Boer goats via Muscular administration of E.colli endotoxin. To study pharmacokinetics of clindamycin in Boer goats of Blood Stagnation Syndrome with a single i.m. dose of [15mg/(kg·b·w)], I had investigation including four groups: the healthy group, the healthy combined Salvia miltiorrhizagroup,the Blood Stagnation Syndrome group and the contrary Syndrome group.Method :24 Boer goats are distributed into 4 groups averagely and randomly. The numbers of female and male are equal in each group. The healthy and the Blood Stagnation group were muscular administrated [15mg/(kg·b·w)]Clindamycin.The healthy combined Salvia miltiorrhiza group and the contrary Syndrome groups were muscular administrated compound injection at same dose (60mg clindamycin and 1.5g Salvia miltiorrhiza per milliliter in compound injection). The concentration of clindamycin in plasma was detected by HPLC. Calculation of pharmacokinetical parameters was performed by 3P97 PK program and further pharmacokinetics analysis was performed by SPSS (12.0) software.Results: (1) the data of the drug concentration-time in the Healthy group, the healthy combined Salvia miltiorrhiza group were fitted to a two-compartment with first-order absorption. The main PK-Parameters of the healthy group were Tp 0.604±0.212h, Cmax2.059±0.219mg/L, AUC 10.376±0.833mg/L ·h, V/F4.574±1.059 L/kg, CL 1.453±0.114 L/kg/h, T1/2 5.26±2.463h and the healthy combined Salvia miltiorrhiza group were Tp 0.503±0.181h, Cmax 3.258±0.947 mg/L, AUC 13.314±2.872 mg/L · h, V/F 3.166±1.326 L/kg, CL 1.164±0.21 L/kg/h, T1/23.512±0.536h.(2) The data of Blood Stagnation Syndrome and the contrary Syndrome group were fitted to one- compartment model with first-order absorption. The main PK-Parameters of the Blood Stagnation Syndrome were Tp0.966±0.173h, Cmax1.366±0.261 mg/L, AUC 7.948±1.85 mg/L · h, V/F 7.386±2.023 L/kg, CL 1.846±0.481 L/kg/h, T1/23.013±1.305h and the contrary Syndrome group were Tp 0.633±0.249 h, Cmax 2.038±0.535 mg/L, AUC 9.991±1.831mg/L · h, V/F 4.042±2.222 L/kg, CL 1.51 ±0.201 L/kg/h,T1/2 3.022±1.135h.Conclusion:(1)TheClindamycin's characteristics of the PK parameters for the health were absorbed fast , reaching peak in short time, distributed extensively (2)The same characteristics of the health combined Salvia miltiorrhiza group hadchanged dramatically. The concentration at peak was increased. Apparent volume of distribution was increased. The elimination of Clindamycin was faster.(3) The same characteristics of the Blood Stagnation group were absorbed fast , reaching peak in longer time, the concentration at peak decreased, apparent volume of distribution decreased, the elimination of Clindamycin faster (4) Salvia miltiorrhiza can rectify changes of the Clindamycin's characteristics of the PK parameters resulted by Blood Stagnation Syndrome. Clindamycin was absorbed quickly and its concentration at peak was increased in the contrary Syndrome group. |
