| The methods of obtaining sweet cherry free from viruses through stem apex culture,thermothetapy combined with stem apex culture and treatment with Ribavirin were studied,12 cultivor sweet cherries as trial materials in this work.The results were showed:(1)survival rate was internal relative with genotype;(2)survival rate was positive correlation with length of stem apex;(3)survival rate was extremely low after thermotherapy ;(4)survival rate was affected by concentration and time of treatment with Ribavirin and the effect of concentration was more visible than that of time.The virus detection results for PNRSV and PDV by ELISA and RNA slide hybridization were indicated:cultivars R-C,A31,W-1,W-2,1-1,KN,S,Y,D were uninfected by PNRSV and PDV,cultivars A30,07,01 were infected by PNRSV only and parts of these three cultivars after virus-elimination were virus-free.The results were showed:(1)obtaining sweet cherry free from PNRSV through stem apex culture was viable,virus-free rate was negative correlative with length of stem apex;(2)virus-free rate was increased by multi-stem apex culture ;(3)best of  these three methods was  thermothetapy combined with stem apex culture,and maximum virus-free rate was100%;(4)Ribavirin was ineffective to eliminate PNRSV in sweet cherry in this study. |
