(?)is studyr BALB/c mice and piglets were immunized with pcDNA-PRRSV-0RF5 DNA, cine of Sichuan isolate of PRRSV-SC2, and BALB/c mice were immunized with pcDNA (?)RRSV-ORF5 DNA vaccine and pig interleukin 2(IL-2) eukaryon expression plas(?) by the routes of co-injection. Fluorescence Activated Cell Sorter (FACS), T (?)te proliferation test(MTT) and indirect ELISA methods were employed (?)ely to detect cellular and humoral immune responses of BALB/c mice and piglets (?) with pcDNA-PRRSV-ORF5 DNA vaccine.(?)B/c mice were immunized with pcDNA-PRRSV-ORF5 DNA vaccine by (?) routes of gene gun bombardment and intramuscular injection. Fluorescence (?)Cell Sorter (FACS) was used to detect the number of CD4~+ and CD8~+ T (?) tes, T lymphocyte proliferation test(MTT) and indirect ELISA methods were (?) respectively to detect the proliferation of peripheral T lymphocyte and (?)V antibody. Peripheral T lymphocytes responses of the experiment group and (?) ol group to ConA were significantly different (P<0.01). In contrast with the (?)oup, the number of CD4~+ T lymphocyte 7 days after vaccination and CD8~+ T (?)te 28 days after vaccination of the experiment group was significantly higher. (?) of specific IgG of the experiment group were higher than that of the control (?)e results demonstrated that pcDNA-PRRSV-ORF5 DNA vaccine could induce (?)oduce good humoral and cellular immune responses. The titers of specific IgG of (?) vaccinated by gene gun bombardment were higher than that by intramuscular(?)B/c mice were immunized with 50,100,200μg of pcDNA-PRRSV-ORF5 DNA (?)spectively by the route of intramuscular injection, Fluorescence Activated Cell (?) ACS) was used to detect the number of CD4~+ and CD8~+ T lymphocytes. T (?)te proliferation test(MTT) and indirect ELISA methods were employed (?)ly to detect the proliferation of peripheral T lymphocyte and anti-PRRSV (?)he results showed ConA response of T-lymphocytes in blood was highsignificant of differences between all experiment groups and the control(P<0.01). The number of CD4+ T-lymphocytes of experiment groups was significantly higher than that of the control 7 days after vaccination. The number of CD8+ T-lymphocytes of experiment groups was higher than that of the control group 28 days after vaccination. Humoral immune response were detected by indirect ELISA. Mice immunized with higher dose (200ug) of DNA vaccine demonstrated higher humoral and cellular immune responses than mice immunized with lower dose(100, 50ug) of DNA vaccine. The results demonstrated that pcDNA-PRRSV-ORF5 DNA vaccine could induce good humoral and cellular immune responses and it showed some dose-dependent character.BALB/c mice were immunized with pcDNA-PRRSV-ORF5 DNA vaccine and pig interleukin 2(IL-2) eukaryon expression plasmid by the routes of co-injection and DNA vaccine injection alone respectively, with PBS and pcDNA3.1(+) as controls. Fluorescence Activated Cell Sorter (FACS) was used to detect the number of CD4+ and CD8+ T lymphocytes, T lymphocyte proliferation test(MTT) and indirect ELISA methods were employed respectively to detect the proliferation of peripheral T lymphocyte and anti-PRRSV antibody. The experiment group vaccinated with DNA vaccine and pig IL-2 eukaryon expression plasmid co-injection showed higher humoral and cellular immune responses than that vaccinated with DNA vaccine alone. The results demonstrated that pig IL-2 eukaryon expression plasmid was good adjuvant which could significantly enhance the immunogenicityofpcDNA-PRRSV-ORF5 DNA vaccine.Piglets were immunized with pcDNA-PRRSV-ORF5 DNA vaccine by the route of intramuscular injection. The results showed ConA response of T-lymphocytes in blood was high significant of differences between experiment groups and the control(PO.Ol). The number of CD4+ T-lymphocytes of experiment groups was significantly higher than that of the control 7 days after vaccination. The number of CD8+ T-lymphocytes of experiment groups was higher than that of the control group 15 to 60 days after vaccination. Humoral immune response were detected by indirect ELISA. The titers of specific IgG of the experiment group were higher than that of the control group(P<0.01).The results indicated that pcDNA-PRRSV-ORF5 DNA vaccine could induce piglets to produce good humoral and cellular immune responses, and humoral immune response persisted for more than 135 days.
