Pearly Mussel include Hyriopsis cumingii Cristaria plicata and Anodonto woodlana, spread widely in eastern, southern and the center of china. There are abundant of pearly mussels in our country, the research of it constitutes of morphilogic ecology biochemistry cell biology molecular biology and so on. The researching working about clam localized in important species in the sea, the pearl mussels in freshwater weren't studied in though they are much important in today's life along with the expanding scales. With the large quantity catching and density culturing, the genetic diversity of pearl mussels were destroyed in different degree, the resources deduces year and year. Working about the genetic diversity of three species of pearl mussels in freshwater, will benefit protesting the resource of pearl mussels, and provide the academic reason for breeding and output in great large scale. From the Nov. in 2002 to the Nov. in 2003, the author studied the resources identification the relations! the genetic diversity of the three species of pearl mussels in freshwater by AFLP in Aquacultural genetic engineering lab in HUNAU. The main results from AFLP fingerprintings and cluster analysis indicated: (1)Meliorating the routine CTAB and SDS methods to get the DNA , we integrate the two methods and optimized the condition, after electrophoresis by AFLP with the DNA, we found the fingerprint is clear; (2)Adopting eight primer pairs to analysis the relationship in genus and among genus, in the analysis among genus, there are 565 amplified sites with 504 polymorphic sites, polymorphism proportion is 89.20%, and with 10.80% differentiation rate. In the analysis of Hyriopsis cumingii, we got 245 sites and including 61 polymorphic sites, the polymorphism proportion is 24.87%; There are 251 sites including 51 polymorphic sites in Cristaria plicata, the polymorphism proportion is 20.40%; 191 sites including 30 polymorphic sites in Anodonto woodlana, the polymorphism proportion is 15.85%. The results show that the 8 pair of primers fit to analysis the population resources of freshwater pearl mussels by AFLP. ?For polygenetic relationship among mussel, genetic distances were calculated and used for constructing a dendragram by UPGMA clustering analysis. The results suggest that the relationship of Hyriopsis cumingii and Cristaria plicata. is more approximation. The similarity coefficient is between 0.5631 to 0.8526 in Hyriopsis cumingii and 0.6724-0.8751 in Cristaria plicata, while the coefficience is between 0.7865-0.8960 in Anodonto woodlana, this means that the Anodonto woodlana has the most similarity meanwhile the Hyriopsis cumingii has the most luxuriant genetic diversity.
