Prunus sargentill is a species of hekistotherm which originated from Hokkaido of Japan and with a high visual value .In 1972, this plant was presented by the later Japan prime minister for resuming the diplomatic relations china and Japan . Up to now, the ages of the trees have been over 30 years, but the quantity is reducing gradually. The development of them is restricted by the unsuitable climate and soil, which shows in the difficulties in bourgeon and low germination rate of seeds.In this dissertation,with the comparison of seed biological characteristics ,the biological test of germination inhibitory substance in different sits of seed and the separation and test of phenolic , ocidity and alkalinity substances and zsoenzyme zynogram in seeds which with different GA3 concentrations . The results showed that the main reasons of the seed dormancy of Prunus sargentill were the machnical resistance of testa and inhibitory substances, not the permeability of gas and water. Through using the GC-MS technology, 13 kinds of endogenic germinating inhibitors in embryo were examined and the result indicated that the phenolic compounds were the main inhibitors. In the zymogram of isoenzymes of peroxidase and α-amylase, some special bands possibly related the seed dormancy of Prunus sargentill were found.Through the research of the dormancy mechanism of seeds of Prunus sargentill, an effective method of getting rid of the dormancy of the seed was found as follows: firstly, soak the seeds in the concentrated for 40 min vitriol; secondly soak then in 400mg/L GA3 for 24h; finally, accelerate the germination through stacked volume by mixing the seeds and sand in a proportion of 1:5 under 3-5 ℃ for one month.
