Presently, the improvement in poultry breeding has been made. However, negativeeffects have appeared accompanied with high intensity selection..Problems neededresolved by poultry industry in China, exist in controlling excessive fatnessaccumulation that affects chicken. carcass quality.To resove these problems, one mainmethod to select fat and flavour traits is using marker-assisted selection (MAS) basedon molecular genetics.In this study, AMPD1 gene and A-FABP gene were studied ascandidates for IMP concentratioin and fat traits in chicken。 AMP-deaminase1(AMPD1) catalyzes the irreversible hydrolysis of AMP to IMPand ammonia being the integral enzyme of purine nucleotide cycle. This study wasdesigned to investigate the effect of AMPD1 gene on chichen IMP concentratioin.Thefourth generation population of divergent selection Yinxingbai broiler ,Laihang layerand two Chinese native chicken breeds(Beijing you and Sanhuang huxu)were used inthe research.Body weight and body composition traits were measured in Beijing you at90 days.Primer pb were designed.Polymophisms were detected by PCR-SSCP andDNA sequenceing.The results showed that there was significant diffrence (P<0.01)inthe frequency of genetype among breeds except Sanhuang huxu vs Yinxingbai broilerand Beijing you vs Bailaihang.A/B mutation was found among individule in Beijingyou and One-way ANOVA analysis showed that AA birds had lower(p<0.05) IMP thanAB and BB birds.It implied that AMPD1 gene could be as candidate locus or linked toa major gene to significantly affect IMP concentration in chicken. In the second experiment,Yinxingbai broiler ,Laihang Leyer and two Chinesespecial breeds (Beijing you and Sanhuang huxu) were used to study the effect 0fA-FABP gene on fat traits.The first exonand the first intron of the gene was amplifiedby two pairs of primer(P1 and P2),and then single nucleotide polymorphism(SNPs)were detected by the technique of single strand conformation polymorphism (SSCP)and finally confirmed by sequencing.(1)P1:The results of X2 analysis showed that the distribution of three genotype wasvery different among four breeds (P<0.01).The results of variance analysis showed thatthe birds with BB genotype had a higher abdominal fat ratio and subcutaneous fatthan the birds with other genotype (AA and AB).,and the birds with BB genotype had ahigher intramuscular fat than AB genotype.(2)P2:The results of X2 analysis showed that the distribution of three genotype wassignificant different among four breeds (P<0.01).The results of variance analysis3showed that the birds with AA genotype had a higher abdominal fat ratio than the birdswith AB genotype.,and the birds with BB genotype had a higher leg muscle ratio thanAB and AA genotype. Least square analysis for different arrrayof polymorphyic loci in A-FABP gene andtheir significant test in Beijing Oil Chick showed that the traits besides leg muscle ratiowere dirrrent among diffren genotype. (p<0.05).It implied that A-FABP gene could be acandidate locus of linked to a major gene to significantly affect fat in chicken.
