| Three cattle were used for cultivating the Calculus Bovis in the artificial gallbladder, from which the drained bile was obtained. The gallbladder bile in cattle was from slaughterhouse. The mixture of the drained bile and the gallbladder bile were utilized to cultivate the Calculus Bovis in generator. It were studied the effects of the temperature and the content of Ca2+,Mg2+,sodium bilirubinate and musin in bile on the ECB and the main ingredients of bile. The results as follows:1.It was favourable to forming sediments of calcium bilirubinate to increase the reaction temperature, the optimum temperature was at 55℃.2.The colour of bile changed from grass green to yellow colour after reacting about 20 hours in the generator, and the viscosity and PH value went slowly.3.E.coli were broken into pieces, the semifinished product of ?-G,by ultrasonic wave, which were used to cultivate the ECB in generator at 55℃,with 40 drops of bile per miniute and 2500ml bile used in each test.(1) Adding Ca2+ into generators, bilirubin precipitates in bile obviously increased (P<0.01), and the optimum concentration of Ca2+ was 5mg/dl.(2) Adding Mg2+ into generators, it had no significant influence on the quality and quantity of ECB, and the main ingredients of the bile (P>0.05).(3) Adding simultaneously both Ca2+ and Mg2+ into generators, it might significantly accelerate bilirubin precipitating in bile (P<0.01). The concentration of bilirubin and musin in bile dropped obviously, although cholic acid and Mg2+ went down slightly (P>0.05).(4) Sodium bilirubinate might increase the bilirubin change rate from conjugated bilirubin to unconjugated bilirubin, and the content of Ca2+ and musin in generators descended smoothly, while other ingredients of bile with slight changes (P>0.05).(5) While bile with 15mg/dl Ca2+ and 15mg/dl Mg2+, and adding sodium bilirubinate into generators,both the output of the ECB and the content of bilirubin,musin or Ca2+ decreased obviously after reaction (P<0.01).(6) While bile with 15mg/dl Ca2+ and 15mg/dl Mg2+ and enhancing the concentration of musin, bilirubin,Ca2+ and musin contents in bile had significant changes(P<0.05),but had no changes as musin surpassing a certain concentration,which made the content of cholic acid and Mg2+ in bile descend smoothly and slightly(P>0.05).4.There were 30.290g ECB obtained in the experiment, with 33 percent ECB containing over 35%bilirubin. The output of ECB was about 3.029g/10000ml bile.5.The pharmacological actions of ECB were similar to that of the natural Calculus Bovis and the Cultured Calculus Bovis.For example, the ECB might resist the rats fever induced by 2,4—dinitrophenol, and might decrease the normal blood pressure in rabbits. |
