142 maize (Zea mays. L.) inbred lines and hybrids widely used or being extended in Sichuan were analysized by SSR(Simple Sequence Repeat). A highly effective, accurate and reliable identification system was set up. DNA fingerprints database of maize inbred lines and hybrids widely used in Sichuan was constructed. Primers with high polymorphism were screened. The study showed that SSR technique was feasible and effective to identify seed authenticity and purity of maize.The main results were summarized as follows:1. SSR technique was feasible to identify seed authenticity of maize inbred lines.77 inbred lines widely used in Sichuan were analysized by SSR. Twenty out of ninety-four pairs of SSR primers were selected,which gave stable and polymorphic amplification profiles. The DNA fingerpints were constructed by the fingerprints combination of six pairs of primers of 233,250,265,277,286,290,which can discriminate the different inbred lines separately. What's more, fifteen inbred lines, such as "698-8", "YA3729", can be discriminated from others by only one pair of primer. Sib-inbred lines closely connected within genetic backgrounds can also be differentiated effectively.2. SSR technique was also feasible to identify seed authenticity and purity of maize hybrids. Analysis on sixty-five maize hybrids widely used or being extended in Sichuan was carried out by SSR. Twenty-two out of ninety-four pairs of SSR primers were screened, which gave high polymorphisms,seven from which can discriminate 65 maize hybrids from each other,which established the DNA fingerprints of the materials. Moreover, 8 maize hybrids, such as "Chuandan 9 ", "Chuandan 26",can be discriminated from other hybrids by only one pair of primer.3. The database of SSR fingerprintings of 77 maize inbred lines and maize hybrids was established. We also set up the automatic identification system of maize DNA fingerprints. The system can recognize the fingerprints, check the fingerprints automatically and identify the fingerprints. So we can apply it to identify seed authenticity and purity of maize.4. The identification system based on SSR molecular marker can differientiate maize hybrids and their parents clearly. The fingerprints of the hybrids appeared to be complement of the parents, which can identify the purity of maize hybrids precisely and quickly.lt was proved to be accurate and dependable by artificial blending test. The purities and authenticities of maize hybrids or inbred lines offered by seed companies inside and outside Sichuan Province, were analysized without problem.The practical application proved that the system was accurate and reliable.
