Eucommia Ulmoiudes Olive, is a special local product in our country and it is a kind of traditional and rare medicinal herbs. It is much valuable. The paper reports something about it. The callus is induced and proliferated using young stem as explant, which comes out at early spring. In the paper, author discussed the inducing effect and the proliferation effect by using different kinds of hormone and different physical-chemical factors. The growth-circle and some physiology-biochemical norm are illustrated. At the same time, the tendencies of Eucommia Ulmoide Gum(EUG) and Aucubin during the growth-circle are analysed. The paper provides scientific basis for cell culture and secondary metabolism matters yield.The following are results:1. The effects of induction and the proliferation which mediums are applied different kinds of hormones or different concentration hormone are different. In the long run, medium MS+NAA1.0mg/L is the optimum medium for callus induction, and medium MS+NAAo.5mg/L+6-BAo.5mg/L+KT1.0mg/L is optimum for callus proliferation.2. The effects of different physical-chemical factors are different. When the concentration of glucode is 30g/L, the effect of callus proliferation is the best of all.3. Medium MS does good to callus proliferation.4. LH does harmful to callus proliferation.5. The growth-circle of callus is 24 days and the growth-curve looks like "S". The growth -circle could be divided 3 parts: Delay(0-9d), Index(9-18d), Steady(18-24d).6. During the growth -circle, the tendency soluble protein contents, SOD actives and POD actives is different during 3 parts. They show some physiology-biochemical norm during callus proliferation and cell actives. It is different between the growth-curve of callus cultured brightly and that of callus cultured dark.7. Secondary metabolism matters accumulate during the Steady(18-24d). The content of Eucommia Ulmoide Gum(EUG) is 2.35% and overpasses 30% of the content of maternal plant ,and the date for harvest yield is about the 21sday. The content of Aucubin is 0.0831%, overpasses the 60% of the content of maternal plant. The date for harvest yield is about the 24th day. Both of that are much more when callus is cultured brightly.
