Two Ribozyme genes targeting negative strand RNA of PLRV replicase gene with two cleavage and single cleavage sites , which were constructed into binary vector plasmid pROKllrespectively The recombinant plasmid pRRz and pRDRz were transfered into Agrobacterium tumefaciens LBA4404 by tn-parental conjugation Potato cultivar Desiree and Wumeng 601 were transformed with single Ribozyme gene and Desiree was transformed with double Ribozyme gene The transgenetic plant.le.ts were regenerated on root inducing medium . PCR analysis showed that two nibozyme genes were intergrated into genome of potatoes respectively ,and Northern hybridization tests showed that nibozyme RNA was found.
