| The Fasciola hepatica Lmneuus, 1758 and Fasciola g~gantica Cobbold, 1855 belong to Fasciola Linnaeus ,1758 of Fasciolidae Raillet , I 895.Both of them infect the ruminant and human ,so the loss caused by them is enormous .However the difference of pathogenesis ,the immunity mechanism of host ,and other aspects of them is still not well-known just because of the similar shapes of them ,both the worm1s body and eggs. We have collected some Fasciola in Jingning county in Jiangsu Province, in which ,some worms resemble the Fasciola hepaica, others is hard to be identified by shoulders .ln order to assure the taxonomy status of a Fasciola ,we analysed the morphology and polymorphism of genomics DNA of those Fasciola. The result is as following: (1). The shoulders .. intestinal caeca width.. length and length/width of these worms (FsspThJ) was studied., with control of Fasciola hepatica from Heilongjiang province ,Guangdong province ,Neimeng in China. , England ,and German; the Fasciola gigantica. from Guangxi province. The result is confusing for the data and indicate that Fasciola FsspJN may be a Fasciola indica or a Fasciola glgantica without typical morphology (2) .ln RAPD analysis ,Ten primers are applied to detect the difference among Fasciola hepatica and Fasciola gigantica from China, England and German. The amplification results show that eight out often primers can produce DNA fingerprint. After cluster analysis with SPSS software, the RAPD results coincide with the results by morphology. The assay also indicate that the Fasciola from iiangning county in China consist of Fasciola hepatica with identical morphology and Fasciola gigantica without identical morphology. Additionally ,the extraction of DNA, optimization of RAPD is also discussed. (3) Whether the RAPD patterns can be repeated is often discussed. In our assay ,the effect of Taq enzyme from the same factory with different lot.No ,PCR cycler from different factory on RAPD pattern .The result indicated that the PCR cycler from different factory show some effect on the RAPD patterns, while the effect of the Taq enzyme from the same factory with different lot.No on RAPD patterns were not detected. Additionally, in order to distinguish egg of the Fischoederius elongatus, Fa.sciolo hepatica, Schistosoma japonicum worms ,we succeed in finding three RAPD patterns ,which can be made into probes for identification. |
