Kongyu 131 Heilongjiang province, was introduced from Japan, adapted to the second accumulated temperature of the cold area dry area and the third accumulated temperature area cultivated japonica rice varieties, the rice varieties with cold tolerance, high yield and quality characteristics, so in recent years has been the Heilongjiang ricethe largest varieties of cultivated area. Study by Basta resistance screening, indoor insect identification, PCR assay, Southern blot, RT-PCR detection, Bt protein quantitative detection and agronomic characteristics of examining and other steps, the proceeds of transgenic plants for insect resistance evaluation and molecular identification, access to value of transgenic plants in the following experiments described in to facilitate the record after the identification of transgenic plants containing crylC* and bar was defined for HDL The main findings are as follows:1. In 43 HD1 family examined by basta, All the result of families can resistance basta. select in all resistance familes examined by PCR, the result of 32 families including two genes of cry1C* and bar genes, and the other families containing bar gene2. Select in 11 HD1 plants which contain cry1C*and bar genes came from 32 HD1 plants and 1 plants containing the bar gene independent plants indoor insect-resistance identification, and select ck by Kongyu 131,the result of 6 plants which has insect-resistance.it included HD1-12. HD1-15, HD1-17, HD1-24, HD1-29 and HD1-42.3. Select in 4 HD1 plants of healthy plant from 6 insect resistance families, besides select in 1 plant bar gene independent through Southern blot detection, the result of which contain copy number between 1 or 2, in plants which contained one copy in HD1-12 and HD1-15, and 2 plants for one copy in HD1-29 and HD1-42; and 1 plant containing the bar gene as a single copy integration.4 At the same time, in 4 HD1 plants which is the same with step 3, and grown healthy and insect resistance exceeding in semi-quantitative RT-PCR detection and quantitative detection of Bt protein. The result of which contained crylC* transcription and translation of the highest levels was marked HD1-29. It’s Cry1C* protein content of 3.30 ± 0.02μg in 1g leaf fresh weight.5. Select grown healthy and insect resistance 4 plants of agronomic traits detection which contained HD1-12, HD1-15,HD1-29, HD1-42 and wild-plant.the result of agronomic traits which has not difference between trans-genetice and wild plant kongyu131 for ck,the result of HD1-29 plants and ck has not differential,the high expression HD1-29 has a strongly potential in commodity.
